PAI-1 spielt eine wichtige Rolle in Tumorinvasion und Metastasis, vor allem bei Brustkrebs. Außerdem werden gesteigerte Werte mit Adipositas, dem metabolischem Syndrom und zahlreichen anderen Erkrankungen in Verbindung gebracht. PAI-1 hat nicht nur die Funktion eines Inhibitors, sondern ist unter
anderem auch an der Zellmigration beteiligt.
In dieser Arbeit werden die Auswirkungen von Zink auf die PAI-1 Expression in MCF-7 Zellen sowie eine mögliche Verbindung mit dem MAPK-Pathway näher betrachtet. Dieser Zusammenhang ist interessant, da sowohl PAI-1 als auch die MAPKinasen pERK 1/2 und p-p38 von Zink stimuliert werden und alle drei
Komponenten für Krebsprogression und Metastasis entscheidend sein können. Die Untersuchungen zeigten eine signifikante Stimulierung der Aktivität von pERK 1 und 2, p-p38 und der Expression von PAI-1 durch Zink in einer Konzentration von 100μM. Die PAI-1 Stimulierung konnte nur durch die
Inhibierung von pERK 1/2 rückgängig gemacht werden. p-p38 zeigte hingegen keine Bedeutung für die Zink-induzierte Stimulierung von PAI-1. Des Weiteren wurde untersucht, ab welcher Konzentration von Zink die Aktivierung der
MAPKinasen und von PAI-1 beginnt, um die physiologische Relevanz genauer zu beleuchten. Es ist so, dass sowohl PAI-1 als auch pERK 1/2 und p-p38 ab einer Konzentration von 100μM Zink eindeutig stimuliert werden. Außerdem wurde das Adhäsions- und Migrationsverhalten von MCF-7 Zellen nach
Inkubation mit Zink und den Inhibitoren von pERK 1/2 und p-p38 untersucht. Der MTT-Test zeigte keine Auswirkungen der MAPKinasen auf die Proliferation, es erfolgte jedoch eine signifikante Abnahme durch Zink in einer Konzentration
von 150μM. Die Zellablösung, die mittels Kristallviolett-Test ermittelt wurde, lieferte kein eindeutiges Ergebnis. Verschiedene Zinkkonzentrationen zwischen 10 und 200μM zeigten keinen Einfluss auf die Migration von MCF-7 Zellen. Diese Versuche wurden durchgeführt, da Proliferation, Adhäsion und Migration
wichtige Rollen in der Krebsprogression spielen. Abschließend wurde auch die Gelatinase-Aktivität von MMP-2 und MMP-9 gemessen, da auch diese große Bedeutung in Brustkrebs haben. Es konnte allerdings keine messbare Aktivität der MMPs festgestellt werden, was auch mit der Literatur übereinstimmt.
Zusammenfassend kann gesagt werden, dass die Zink-induzierte Stimulierung von PAI-1 in MCF-7 Zellen über den ERK-Pathway reguliert wird. Diese Erkenntnis könnte Ansätze für weitere Untersuchungen liefern, die zum besseren Verstehen der komplexen Zusammenhänge in Brustkrebs beitragen.
Ebenso könnten diese Versuche auf weitere Zelllinien ausgeweitet werden, da PAI-1 nicht nur in Brustkrebs eine wichtige Rolle spielt.
Abstract

PAI-1 plays an important role in tumor progression and metastasis, especially in breast cancer. Increased levels are also associated with adipositas, the metabolic syndrom and many other diseases. PAI-1 does not only function as an inhibitor, but is also involved in the process of cell migration.
In this study, the effect of Zinc on the expression of PAI-1 in MCF-7 cells was investigated, as well as the involvement of the MAPK-Pathway. This link is of interest, because PAI-1 as well as the MAPKs pERK 1/2 and p-p38 could be stimulated by Zinc and all three components could be crucial for cancer progression and metastasis. Examinations of the activity of pERK 1/2, p-p38 and the expression of PAI-1 show a significant stimulation by Zinc applied in a concentration of 100μM. The PAI-1 stimulation could only be reversed by inhibition of the pERK 1/2 pathway. p-p38 didn’t show any relevance for the Zinc-induced stimulation of PAI-1. This study further examined the concentration of Zinc needed to induce the activation of the MAPKs and PAI-1, to explore the physiological relevance. It could be shown that PAI-1, pERK 1/2 and p-p38 are all significantly stimulated at 100μM ZnCl2. In addition, the adhesion and migration behaviour of MCF-7 cells after incubation with Zinc and the inhibitors of pERK 1/2 and p-p38 were investigated. The MTT-assay showed no effect of the MAPKs on the proliferation rate, but there was a significant decrease in proliferation induced by Zinc in a concentration of 150μM. The cell detachement, which was measured by the crystalviolet-assay, revealed no steady result. Zinc concentrations between 10 and 200μM did not influence the migration behaviour of MCF-7 cells. These experiments were performed, because proliferation, adhesion and migration play important roles in cancer progression. Finally, the activity of the
gelatinases MMP-2 and -9 was measured, due to their great importance in breast cancer. There was no measurable activity of these MMPs, in accordance with literature.
In summary the data suggest that the Zinc-induced stimulation of PAI-1 is regulated by the ERK-Pathway in MCF-7 cells. This result provides a new background for more investigations that could help to understand the complex mechanisms in breast cancer. Moreover these experiments could be extended
to other cell lines, as the role of PAI-1 is not restricted to breast cancer.
