In der vorliegenden Arbeit verwendeten wir kultivierte Fibroblasten von Friedreich’s Ataxie-Patienten und gesunden Kontrollpersonen, um die Wirkung von rekombinantem humanem Erythropoietin auf die Sensivität von Fibroblasten gegenüber Radikale bildenden Oxidantien zu untersuchen. Unser Ziel mit diesem zellbasierten Ansatz für Friedreich’s Ataxie lag darin, aussagekräftige und reproduzierbare Tests für Friedreich’s Ataxie zu finden. Die untersuchten Parameter waren: Beurteilung der Bildung von oxidativem Stress mittels Dichlorodihydrofluorescein-Diazetat (H2-DCF-DA), sowie zwei Methoden zur Bestimmung der metabolischen Aktivität mittels Resazurin und MTT-Test. Dabei wies das DCF-Verfahren eine höhere inter- und intra-Assay Streuung auf, als die Resazurin-Methode und der MTT-Test. Des Weiteren wurden die Effekte von Tert-butylhydroperoxid (TBH) und H2O2 untersucht und gezeigt, dass TBH im Gegensatz zu H2O2 die metabolische Aktivität in FAFib stärker herabsetzte, als in Kontrollzellen. Mit DCF konnte weder durch TBH, noch H2O2 eine Erhöhung im oxidativen Stress detektiert werden. RhuEPO Präinkubation der Zellen hatte keinen statistisch signifikanten Effekt auf den basalen oxidativen Stress und die metabolische Aktivität im DCF-Assay und mit Resazurin, verbesserte aber die metabolische Aktivität im MTT-Test. Bei zellfreien Untersuchungen wurde auch der direkte Einfluss von rhuEPO und den verwendeten Oxidantien/Radikalbildern auf die Hydrolyse und Oxidation von H2-DCF-DA untersucht. Es zeigte sich, dass rhuEPO keine hydrolysierende, aber eine oxidierende Wirkung gegenüber H2-DCF-DA hatte, die zellprotektive Mechanismen induzieren könnte. Abschließend wurde in einer Pilotstudie der Eisengehalt von zwei Zelllinien von Friedreich’s Ataxie-Patienten, sowie von Kontrollfibroblasten untersucht und gezeigt, dass der zellulärer Eisengehalt wahrscheinlich aufgrund des unterschiedlichen genetischen Backgrounds extrem unterschiedlich ist.

We used cultivated fibroblasts from Friedreich’s ataxia patients and healthy controls to investigate the effect of recombinant human erythropoietin on the sensitivity of fibroblasts to radical-forming oxidants. Using this cell-based approach for Friedreich’s ataxia we aimed to find a robust reproducible readout for the effects of a possible treatment for Friedreich’s ataxia. The investigated readouts were: assessment of oxidative stress generation using the probe dichlorodihydrofluorescein-diacetate
(H2-DCF-DA) as well as two methods to assess metabolic activity using resazurin and MTT as probes. We found that the DCF-method had a high inter-and intra-assay variation than the resazurin-assay and the MTT-test. Furthermore the effects of tert-butylhydroperoxide (TBH) and H2O2 were investigated and showed that TBH decreased, in contrast to H2O2, the metabolic activity of FAFib to a higher extent than in control cells. With the DCF-assay no oxidative stress was detectable resulting from incubation with TBH or H2O2. RhuEPO preincubation of the cells had no statistically significant effect on the basal oxidative stress and metabolic activity in the DCF-assay and with resazurin, but improved the metabolic activity in the MTT-test. In addition we investigated the direct influence of rhuEPO on hydrolysis and oxidation of H2-DCF-DA in cell-free studies. RhuEPO showed oxidative, but no hydrolytic properties in the DCF-assay, a property which may be responsible for induction of cytoprotection by rhuEPO. Finally the iron content in two cell lines from Friedreich’s ataxia patients as well as control fibroblasts was investigated and showed that it is very different probably due to the different genetic background.
