Pentadesma grandifolia zählt zur Familie der Clusiaceae und wird in Kamerun traditionell als Medizinalpfanze eingesetzt. Die in dieser Arbeit beschriebene phytochemische Untersuchung der methanolischen Extrakte ihrer Rinde, Wurzeln, Blätter und Früchte umfasst die Isolation von unterschiedlichen Sekundärmetaboliten, die strukturell zu den Triterpenen, Xanthonen und Biflavanoiden gehören. Aus der Rinde und aus den Wurzeln wurden fünf, respektive drei Verbindungen rein isoliert. Aus Blättern und Früchten wurde je eine Verbindung rein gewonnen. Diese isolierten Substanzen wurden als Lupeol (PS3), β-Amyrin (PS4), Betulin (PS5), Lupenone (PF1), 1,6-Dihydroxy-3,7-dimethoxy-2-(3-methylbut-2-enyl)-xanthon (PS1), α-Mangostin (PS2), Allanxanthon C (PL1), 3,8”-Binaringenin (PR1), 3,6”-Binaringenin (PR2), 3,8”-Binaringenin-3”-O-ß-glucosid (PR3) identifiziert. Die vergleichende Analyse der methanolischen Extrakte aus den unterschiedlichen Pflanzenorganen (Rinde, Wurzeln, Blätter, Früchte) mittels HPLC-UV und TLC erlauben die qualitative und quantitative Bestimmung der verschiedenen Substanzgemische. Deren Hauptkomponenten werden dann mit präparativen Methoden (CC, präp. TLC, HPLC-UV, Sephadex-Säule) getrennt und gereinigt. Die molekularen Strukturen der isolierten Reinstoffe wurden mittels 1D NMR, 2D NMR und Massenspektrometrie ermittelt und durch Abgleich mit Daten literaturbekannter Verbindungen aus Datenbanken verifiziert. 3,6”-Binaringenin (PR2) und 3,8”-Binaringenin-3”-O-ß-glucoside (PR3) werden im Rahmen dieser Arbeit erstmalig beschrieben. Die weiteren isolierten Substanzen wurden erstmalig aus P. grandifolia isoliert, sind aber bereits aus anderen Pflanzen beschrieben worden. Einige der isolierten Verbindungen kommen in mehreren Pflanzenorganen (Rinde, Wurzeln, Blätter, Früchte) vor, andere sind organspezifisch. Die organspezifischen Vorkommen wurden mit vergleichender TLC und HPLC-UV analysiert. α-Mangostin (PS2) ist die dominante Verbindung in der Rinde, während 1,6-Dihydroxy-3,7-dimethoxy-2-(3-methylbut-2-enyl)xanthone (PS1), 3,8”-Binaringenin (PR1) und 3,6”-Binaringenin (PR2) als Hauptkomponenten in den Wurzeln identifiziert wurden. Die Quantifizierung wurde durch Kalibrierung mit Vergleichssubstanzen im HPLC-UV durchgeführt. Weiterhin wurde von den Substanzen eine Bioautographie mit Cladosporium sphaerospermum durchgeführt, um deren antifungale Aktivitäten zu überprüfen. Insektizide Wirkungen wurden darüberhinaus in einem Bioassay mit Spodoptera littoralis geprüft. Die vier Verbindungen 1,6-Dihydroxy-3,7-dimethoxy-2-(3-methylbut-2-enyl)xanthone (PS1), α-Mangostin (PS2), 3,8”-Binaringenin (PR1) und 3,6”-Binaringenin (PR2) zeigen antifungale Aktivität, während keine der Verbindungen eine toxische Wirkung gegenüber Spodoptera littoralis aufweist. Darüberhinaus sind einige der isolierten Verbindungen bereits dafür bekannt, dass sie entzündungshemmend oder Zytotoxisch wirken, bzw. eine anti-Malaria Wirksamkeit aufweisen. Ein zusammenspiel dieser Wirkungen stellt eine Begründung für die Nutzung von P. grandifolia in der traditionellen Volksmedizin in Kamerun dar.

The phytochemical study of the methanolic extract of the stem bark, the roots, the leaves and the fruits of Pentadesma grandifolia, a Cameroonian medicinal plant belonging to Clusiaceae family, permitted the isolation of secondary metabolites belonging to three different classes: Triterpenes, xanthones and biflavonoids. Five compounds were isolated from the stem bark, three from the roots, one from the leaves and from the fruits. These compounds were identified as lupeol (PS3), β-amyrin (PS4), betulin (PS5), lupenone (PF1), 1,6-dihydroxy-3,7-dimethoxy-2-(3-methylbut-2-enyl)-xanthone (PS1), α-mangostin (PS2), Allanxanthone C (PL1), 3,8”-binaringenin (PR1), 3,6”-binaringenin (PR2), 3,8”-binaringenin-3”-O-ß-glucoside (PR3). Comparative analysis of the methanolic extract of different organs of the plant (stem bark, roots, leaves, fruits) by HPLC-UV and TLC revealed the presence of different compounds which were isolated by means of chromatographic techniques such as CC, prep.TLC, MPLC, sephadex column. Their structures were elucidated and identified on the basis of spectroscopic techniques such as NMR 1-D, NMR 2-D, MS and by comparison of their spectra data with those available in literature. 3,6”-binaringenin (PR2) and 3,8”-binaringenin-3”-O-ß-glucoside (PR3) were new natural structure found in plant, the other secondary metabolites isolated from P. grandifolia are new reports for the genus, but they were previously isolated from other plant species. Among these compounds isolated, some co-occur in different organs (stem bark, roots, leaves, and fruits) and some are organ-specific. The organ specific accumulation was analyzed by comparative TLC and HPLC-UV. α-Mangostin (PS2) was the major compounds in the stem bark whereas 1,6-Dihydroxy-3,7-dimethoxy-2-(3-methylbut-2-enyl)-xanthone. (PS1), 3,8”-binaringenin (PR1), 3,6”-binaringenin (PR2) were the major compounds of the roots and their quantification in different organ were done through calibration in HPLC-UV. Additionally, the bio-autography using Cladosporium sphaerospermum was performed to test for antifungal activity, and insecticidal activity was tested in a bioassay with Spodoptera littoralis. Four of the compounds isolated [(1,6-Dihydroxy-3,7-dimethoxy-2-(3-methylbut-2-enyl)-xanthone (PS1), α-mangostin (PS2), 3,8”-binaringenin (PR1) and 3,6”-binaringenin (PR2)] possess antifungal activity but none of these compounds isolated have showed toxicity against Spodoptera littoralis. Whatever, other biological activities such as anti-inflammatory, anti-malarial and cytotoxic have been described from xanthones and biflavanones isolated now. They may be related to the use of the plant in folk medicine in Cameroon.
