Lipidhydroperoxide (LOOH) entstehen beim Erhitzen von mehrfach ungesättigten Fettsäuren unter Sauerstoff. Diese Peroxide sind in der Lage weitere Lipidperoxidationsreaktionen in Zellmembranen der Darmepithelzellen hervorzurufen und oxidativen Stress durch die Bildung freier Radikale zu verursachen. Unter diesen Aspekten wurden die nicht-enzymatischen Schutzmechanismen der α-und γ-Tocopherole auf LOOH-Einwirkung in der Karzinomzelllinie HT29 und den Adenomzelllinien LT97-2 und LT97-3 untersucht. Mittels Dichlorofluorescin-FACS konnte durch LOOH-Behandlung eine Erhöhung der H2O2-Bildung bei HT29 und LT97-2 und eine Schutzfunktion der beiden Tocopherole gegenüber weiterer Radikalbildung in der Karzinomzelllinie nachgewiesen werden. Ebenfalls konnte die durch Hämatin-Eisen erhöhte H2O2-Bildung durch α-Tocopherol-Behandlung in HT29 und LT97-3 gehemmt werden. An dieser Schutzwirkung haben nicht nur die chemischen Eigenschaften der Tocopherole sondern auch ihre Auswirkungen auf die zellulären Schutzenzyme Katalase und Superoxiddismutase sowie auf die Expression von entzündungs-assoziierten Genen Anteil. α-Tocopherol-Behandlung zeigte eine Erhöhung beider Enzymaktivitäten und eine Abmilderung der LOOH-induzierten Veränderungen, besonders bei HT29 Zellen. Die LOOH-induzierte erhöhte Expression von COX-2 und VEGF war sowohl in HT29 als auch in LT97-3 Zellen nachweisbar. In diesem Zusammenhang zeigte γ-Tocopherol eine Schutzwirkung vor LOOH bei HT29 und α-Tocopherol bei LT97-3 Zellen. Eine Messung des Prostaglandin E2 im Zellkulturüberstand mittels Elisa bestätigte die Schutzwirkung von γ-Tocopherol-Behandlung in LT97-3 Adenomzellen. In HT29 Kulturen wurde eine Verminderung der LOOH-induzierten VEGF Sekretion in das Medium durch α-Tocopherol beobachtet. Als zusätzliches Entzündungskriterium wurde die Aktivität des Transkriptionsfaktors NF-κB mittels Dual-Luciferase Reporter Assay gemessen. Diese wurde durch LOOH Exposition deutlich erhöht und konnte durch 24h α-Tocopherol-Einwirkung signifikant gehemmt werden. Die Ergebnisse dieser Studie weisen signifikante Schutzwirkungen der α-und γ-Tocopherole im Zusammenhang der Lipidperoxidation und dessen Auswirkungen auf intrazellulärer Ebene sowohl in Adenom- als auch in Karzinomzellen auf. Somit eignen sich diese Antioxidantien für weitere krebspräventive Studien, die allerdings noch durch in vivo Studien gezeigt werden müssen.

Lipidhydroperoxides (LOOH) arise from heating polyunsaturated fatty acids in the presence of oxygen. These peroxides are able to induce further lipidperoxidase reactions in the cell membranes of the colon epithelial cells and to cause oxidative stress through the formation of free radicals. Based on this knowledge the non-enzymatic protective mechanisms of α-und γ-tocopherol were studied by treating the carcinoma cell line HT29 and the adenoma cell lines LT97-2 and LT97-3 with LOOH. Through the use of Dichlorofluorescin-FACS it could be shown that LOOH-induced increases of the H2O2 production in HT29 and LT97-2 cells could be largely reduced by the concomitant addition of both tocopherols. The increase of H2O2-formation caused by hematin-iron could be blocked by α-tocopherol in HT29 und LT97-3 cells. In addition to their direct inactivation of reactive oxygen species tocopherols also induce cellular protective enzymes like Katalase and Superoxiddismutase and have an impact on the expression of inflammation-associated genes. Specifically, α-tocopherol-treatment caused an increase in both enzyme activities and a decline in LOOH-induced changes, especially in HT29 cells. Exposure to LOOH raised expression of COX-2 and VEGF in HT29 and LT97-3 cells. This effect was inhibited by γ-tocopherol in HT29 and by α-tocopherol in LT97-3 cells. Measurement of Prostaglandin E2 in the cell culture supernatant through Elisa confirmed the protective mechanism of γ-tocopherol-treatment in LT97-3 adenoma cells. α-tocopherol also reduced LOOH-induced VEGF secretion into the medium of HT29 cultures. As an additional inflammation marker the activity of the transcription factor NF-κB was measured by Dual-Luciferase Reporter Assay. This activity was increased significantly through LOOH exposition and could be reduced significantly through 24h α-tocopherol-treatment. The results of this study demonstrate distinct protective mechanisms of α- and γ-tocopherols in relation to the lipid peroxidation and its effects on an intracellular basis both in adenoma cells as well as in carcinoma cells. This suggests cancer preventive activities of these antioxidants that need to be further tested in vivo.
