Viele neurodegenerative Erkrankungen sind gekennzeichnet durch Eisenanomalie und den dadurch verursachten oxidativen Stress. Im Rahmen dieser Masterarbeit wurden die Behandlungen mit Dimethyloxalylglycin (DMOG) in den Konzentrationen [0,5mM] sowie [1mM] und/oder Erythropoietin (EPO) in der Konzentration [3,3U/ml] im Zusammenhang mit der Adaption des Eisenstoffwechsels in vitro an Neuroblastomzellen (SH-SY5Y Zellen), als Beispiel für eine organspezifische Wirkung im Gehirn, und an Hepatokarzinomzellen (HepG2 Zellen), als Testmodell für den systemischen Effekt, zeitabhängig nach 2h, 6h und 24h untersucht. Die Wirkung und der Effekt von DMOG und EPO auf die Eisenstoffwechselkomponenten labiler Eisenpool (LIP), zytosolisches Ferritin und mitochondriales Ferritin unterscheiden sich zeit-, dosis- und zellartabhängig. In den Neuroblastomzellen zeigten alle Behandlungen nach 2h eine signifikante Reduktion des LIPs. Nach 6h und 24h wurde ein Anstieg des LIP bei der kombinierten Inkubation mit DMOG und EPO gemessen. DMOG alleine führte weiterhin zu einer LIP Senkung. EPO hatte nach 24h auf den LIP keine Wirkung mehr. Die deutlichsten, reduzierenden Effekte auf die zytosolische Ferritinkonzentration wurden nach 24h ermittelt. Signifikanz konnte dabei bei der Einzelbehandlung mit EPO und in Kombination mit DMOG berechnet werden. Bei der mitochondrialen Ferritinmessung gab es eine signifikante Erhöhung nach der Inkubation mit EPO, DMOG in der niedrigeren Konzentration und nach der kombinierten Behandlung. In den Hepatokarzinomzellen führten alle Behandlung außer EPO nach 24h zu einer Erhöhung des LIP, während bei EPO sich kein Effekt zeigte. Bei Ferritin konnten eine tendenzielle Reduzierung festgestellt werden, die jedoch statistisch nicht signifikant war. Auch beim mitochondrialen Ferritin konnten keine signifikante Änderung der Expression ermittelt werden. Zusammenfassend konnte gezeigt werden, dass es zellspezifische Unterschiede zwischen den Zelllinien SH-SY5Y und HepG2 hinsichtlich der Eisenstoffwechselanpassung nach den Behandlungen mit DMOG und/oder EPO gab. In den Neuroblastomzellen konnte in Bezug auf die Eisenadaption eine mögliche neuroprotektive Wirkung durch Reduzierung des freien Eisens nachgewiesen werden.

Many neurodegenerative diseases are characterized by abnormal iron homeostasis and subsequent oxidative stress. The present in vitro study was performed to analyze the effects of dimethyloxalylglycine (DMOG) in concentrations of [0,5mM] and [1mM] and/or erythropoietin (EPO) in the concentration of [3,3U/ml] on the adaptation of iron metabolism. Neuroblastoma cells (SH-SY5Y cells) were used as a test model for organ-specific effects in the brain and hepatoma cells (HepG2 cells) were used as an example of systemic effects. Cells were treated for 2h, 6h or 24h. The effect of DMOG and EPO on the iron metabolism components labile iron pool (LIP), cytosolic ferritin and mitochondrial ferritin are time-, dose- and cell type-dependent. In SH-SY5Y cells all treatments showed a significant reduction of the LIP after 2h. The simultaneous treatment with DMOG and EPO led to an increase of LIP after an incubation of 6h and 24h. DMOG alone led to a further reduction of LIP. The EPO treatment showed no longer effects after 24h of incubation. The clearest reducing effects on the cytosolic ferritin were determined after 24h. Significance was found after the treatment of EPO alone and in combination with DMOG. Mitochondrial ferritin expression was significantly increased after EPO and DMOG [0,5mM] alone and after simultaneous treatment. In HepG2 cells all treatments except EPO led to an increase in the LIP after 24h. Treatment with EPO showed no effects. A trend towards a reduction in ferritin could be detected, but there was no statistical significance. For mitochondrial ferritin no significant change in expression can be detected either. In conclusion, it was shown that there are cell-specific differences between the cell lines SH-SY5Y and HepG2 in terms of iron metabolism adjustment after the treatment with DMOG and/or EPO. In neuroblastoma cells, a potential neuroprotective effect was demonstrated by the adaption of iron metabolism through the reduction of free iron.
