α-Glucosidase-Hemmer, wie z.B. Acarbose und Miglitol, stellen wichtige Arzneistoffe in der Therapie von Diabetes mellitus Typ II dar. Aufgrund der beeinträchtigenden Nebenwirkungen jener Präparate wird jedoch laufend nach neuen, spezifischen α-Glucosidase-Hemmern natürlichen Ursprungs gesucht. Justicia secunda VAHL wird in Ecuador volksmedizinisch zur Behandlung von Diabetes eingesetzt und stellt somit eine potentielle Quelle α-glucosidasehemmender Verbindungen für die Herstellung neuer blutzuckersenkender Medikamente dar. Das primäre Ziel dieser Arbeit war sowohl die Testung verschiedenster Extrakte jener Pflanze auf α-glucosidasehemmende Wirkung, als auch die anschließende Identifizierung und Strukturaufklärung der aktiven Verbindungen. Da nicht nur Pflanzenmaterial aus Ecuador, sondern auch aus Französisch-Guyana zur Verfügung stand, wurde außerdem ein Materialvergleich bezüglich des phytochemischen Fingerprints und der α-glucosidasehemmenden Aktivität erstellt. Zur Fraktionierung und zur Abtrennung störender Begleitstoffe kam die Säulenchromatographie an Polystyrol zum Einsatz. Zum Nachweis einer möglichen blutzuckersenkenden Aktivität diente der sogenannte α-Glucosidase-Inhibitions-Assay. Dünnschicht-, Hochleistungsdünnschicht- und Hochleistungsflüssigchromatographie kamen zur vergleichenden Analyse sowie zur Charakterisierung und Isolierung aktiver Substanzen zum Einsatz. Die Strukturaufklärung aktiver Verbindungen erfolgte über Massenspektrometrie und Kernresonanzspektroskopie. Zwei Verbindungen namens CD1 und SI1, die Aktivität im α-Glucosidase-Inhibitions-Assay zeigten, konnten identifiziert werden. Bei der einen handelte es sich um das Arylpyrrolidon CD1, welches bereits im Rahmen der Dissertation von DI Barbara Theiler isoliert und strukturell aufgeklärt werden konnte. Die zweite Verbindung namens SI1 stellte einen Kaffeesäureester dar, dessen genaue Struktur aufgrund einer zu geringen Ausbeute zwar noch nicht vollständig aufgeklärt werden konnte. Es gelang jedoch mittels LC-MS und NMR einen ersten Strukturvorschlag zu erstellen.

α-Glucosidase inhibitors, such as acarbose and miglitol, represent important agents in the treatment of type 2 diabetes mellitus. Due to their unpleasant adverse effects, new specific α-glucosidase inhibitors from natural origin are constantly searched for. In Ecuador, Justicia secunda VAHL is traditionally used for the treatment of diabetes, so this plant represents a potential source of α-glucosidase inhibiting compounds to establish new hypoglycaemic agents. The primary aim of this work was the testing of various extracts of this plant on α-glucosidase inhibition and the subsequent identification and structure elucidation of the active compounds. Since not only plant material from Ecuador but also from French Guiana was available, a material-comparison with regard to the phytochemical fingerprint and the α-glucosidase inhibiting activity was performed. For fractionation and for the removal of interfering substances, column chromatography on polystyrol was used. The so-called α-glucosidase-inhibition-assay was used to detect a possible hypoglycaemic activity. Thin-layer, high performance thin-layer and high performance liquid chromatography were used for comparative analysis as well as for the characterization and isolation of active compounds. Structure elucidation of active compounds was carried out by mass spectrometry and NMR spectroscopy. Finally, two compounds which showed activity in the α-glucosidase-inhibition-assay, could be identified. One of those was the arylpyrrolidone CD1, which has already been isolated and structurally elucidated as part of the PhD thesis of DI Barbara Theiler. The second compound, termed SI1, represents a caffeic acid ester, but its yield was too low for a full elucidation of the molecular structure. However, it succeeded to create a first structural proposal by LC-MS and NMR.
