Ein grosser Teil des Genoms in Fliegen und Wirbeltieren wird in lange, nicht-kodierende RNAs (lncRNAs) transkribiert. Viele dieser lncRNAs stammen von DNA Elementen, welche von Polycomb Group (PcG) und Trithorax Group (TrxG) Proteinen gebunden werden. Diese Elemente werden Polycomb/Trithorax Response Elements (PREs) genannt. Das vestigial (vg) PRE in Drosophila wird auf beiden Strängen der DNA in lncRNAs transkribiert. Die Expression dieser lncRNAs ist während der Entwicklung in hohem Masse reguliert. Interessanterweise korreliert die Expression der ‘reverse lncRNA’ mit der vg mRNA in Embryos, aber die ‘forward lncRNA’ ist in Zellen der Larve exprimiert, in denen das vg Gen ausgeschaltet ist.
Hier zeige ich, dass die transkriptionelle Aktivität des vg PRE in Embryos mit der Reporter Repression in adulten Fliegen korreliert und die Expression der vg lncRNAs von den PcG und TrxG Proteinen abhängig ist. Die Expression der ‘reversen lncRNA’ von einem ektopischen Lokus führt zur Aktivierung des endogenen vg PRE Lokus in Embryos und die Überexpression der ‘forward lncRNA’ führt zur Reprimierung des vg Gen in sogenannten Flügelscheiben in Larven. Eine Analyse des gesamten Transkriptoms zeigt, dass die ubiquitäre Überexpression der ‘forward’ oder ‘reverse’ lncRNA zur De-regulierung von Genen spezifisch für jede RNA führt.
Ich habe beobachtet, dass in vitro beide lncRNAs die Histon Methyltransferaseaktivität des PcG Komplex PRC2 gegenüber dem Lysin 27 des Histons H3 hemmen. Hingegen bindet nur die ‘reverse’ lncRNA das PRC2 Protein E(Z) in Drosophila S2 Zellen. Chromatin-Immunopräzipitation von E(Z) und H3K27me3 hat gezeigt, dass die ‘reverse’ lncRNA, aber nicht die ‘forward’ lncRNA, lokale PRC2 Proteine vom Chromatin verdrängt und die enzymatische Aktivität in vivo inhibiert. Das deutet darauf hin, dass in vivo die Interaktion von PRC2 mit lncRNAs spezifisch reguliert wird und dies erlaubt spezifische und locale Inhibierung von PRC2. Zusammengefasst präsentiere ich hier eine Arbeit, die spezifische Funktionen für vg lncRNAs, die von den verschiedenen Strängen des Elements transkribiert werden, aufzeigt und dass diese RNAs möglicherweise wichtige Regulatoren für die PcG Funktion in vivo sind.

Large portions of the genome are transcribed into noncoding RNAs (ncRNAs) both in flies and vertebrates. Many of these RNAs are transcribed from DNA elements that are bound by Polycomb (PcG) and Trithorax (TrxG) group of proteins. The Drosophila vestigial (vg) Polycomb/Trithorax Response Element (PRE) is transcribed bi-directionally into long ncRNAs (lncRNAs). The expression of these lncRNAs is highly regulated during development. Strikingly, the expression of the reverse lncRNA correlates with vg gene expression in embryos, whereas the forward lncRNA is expressed in larval tissues specifically in cells, in which the vg gene is repressed.
Here, I show that transcriptional activity at the transgenic vg PRE in embryos correlates with reporter silencing in adult flies and that the expression of the vg PRE transcripts is dependent on both PcG and TrxG proteins.
Furthermore, expression of the reverse lncRNA from an ectopic locus leads to activation of the endogenous vg PRE in embryos and over-expression of the forward strand leads to vg silencing in larval wing discs. A genome-wide analysis shows that ubiquitous over-expression of the forward or reverse lncRNA leads to de-regulation of genes specific for each strand.
In vitro, both lncRNAs inhibit PRC2 histone methyltransferase activity, but that only the reverse strand interacts with the PcG protein E(Z) in S2 cells. Chromatin-immunoprecipitation analysis of E(Z) and H3K27me3 demonstrates that the reverse strand evicts local PRC2 from chromatin and inhibits its enzymatic activity in vivo, suggesting that in vivo the interaction of PRC2 with lncRNAs is specifically regulated and that this allows specific and local inhibition of PRC2.
In conclusion, the work presented here demonstrates specific functions for each vg lncRNAs and that they might be important regulators of PcG function in vivo.
