Entzündungen stellen oft Vorstufen von malignen Prozessen dar und begünstigen Wachstum und Ausbreitung von Tumorzellen. Daher ist Pluchea odorata (L.) Cass., eine Pflanze, die die Maya gegen schwere Entzündungen einsetzen, von Gridling et al. auf eine mögliche anti-tumorale Wirkung untersucht worden. Dabei zeigte der Dichlormethan (DCM)- Extrakt ein starkes antineoplastisches Potential in HL-60 und MCF-7 Zellen. Durch Bioassay geleitete Fraktionierung werden aktive Substanzen isoliert und strukturell identifiziert. Das Ziel dieser Arbeit war, Verbindungen aus bisher noch nicht untersuchten Fraktionen des DCM-Extraktes zu isolieren, die für weitere pharmakologische Untersuchungen verwendet werden können. Mit Hilfe von Säulenchromatographie, Gegenstromverteilungschromatographie und Festphasenextraktion wurden 9 Unterfraktionen des DCM-Extraktes weiter aufgetrennt. Der Fortschritt dieser Analysen wurde mittels Dünnschichtchromatographie verfolgt; acht Substanzen waren rein darstellbar und konnten durch spektroskopische Analysen identifiziert werden: Der Fettalkohol 3,5,16-Trimethyl-heptadec-2-en-ol, die ubiquitären Triterpenalkohole alpha- und beta- Amyrin und das Eudesman Kongol wurden aus sehr lipophilen Fraktionen gewonnen. Aus etwas hydrophileren Fraktionen konnten zwei strukturell sehr ähnliche Sesquiterpene vom Eudesmantyp isoliert werden: (1S,2R,4aR,8aR)-[1-(Hydroxy)-7-(1-hydroxy-1-methylethyl)-1,4a-dimethyl-1,2,3,4,4a,5,6,8a-octahydro-6-oxo-2-naphthalenyl]-2,3-dihydroxy-2-methyl-butansäureester und (1S,2R,4aR,8aR)-[1-(Hydroxy)-7-(1-hydroxy-1-methylethyl)-1,4a-dimethyl-1,2,3,4,4a,5,6,8a-octahydro-6-oxo-2-naphthalenyl]-2,3-dimethyl-oxirancarbonsäureester. Strukturell ähnliche Plucheinol-Derivate sind weit verbreitet in der Gattung Pluchea. In polareren Fraktionen dieses Extraktes waren auch Stigmast-5,22-dien-3-O-beta-D-glucopyranosid und die Chlorophyllabbauprodukte Pheophorbid A und Hydroxy-Pheophorbid A enthalten. Ein Vergleich mit der Literatur hat gezeigt, dass manche Eudesmanolide eine entzündungshemmende und antineoplastische Wirkung aufweisen und somit möglicherweise zur zytotoxischen Aktivität dieser Pflanze auf verschiedene Krebszellen beitragen.

Inflammation often leads to malignant processes and favors growth and proliferation of tumor cells. Therefore, Pluchea odorata (L.) Cass., a plant used against inflammations in Maya medicine, has been examined by Gridling et al. It could be demonstrated that the dichloromethane- (DCM) extract shows strong anti neoplastic effects in HL-60 and MCF-7 cells. Through bioassay guided fractionation active substances are isolated and structurally identified. The aim of this work was to isolate substances of fractions from the DCM-extract for further pharmacological investigations. Using column chromatography, counter current chromatography and solid phase extraction, 9 subfractions of the DCM-extract were separated. The progress was monitored by thin layer chromatography. Fractionation resulted in eight compounds, structure elucidation was performed by spectroscopic methods: The fatty alcohol 3,5,1,-trimethyl-heptadec-2-en-ol, the ubiquitious triterpene alcohols alpha- and beta-amyrine and the eudesmane kongol were isolated from very lipophilic fractions. Two structurally very similar sesquiterpenes with an eudesmane scaffold were isolated: (1S,2R,4aR,8aR)-[1-(Hydroxy)-7-(1-hydroxy-1-methylethyl)-1,4a-dimethyl-1,2,3,4,4a,5,6,8a-octahydro-6-oxo-2-naphthalenyl]-2,3-dihydroxy-2-methyl-butanoic acid ester and (1S,2R,4aR,8aR)-[1-(Hydroxy)-1,2,3,4,4a,5,6,8a-octahydro-7-(1-hydroxy-1-methylethyl)-1,4a-dimethyl-6-oxo-2-naphthalenyl]-2,3-dimethyl-oxiranecarboxylic acid ester from more hydrophilic fractions. Structurally similar plucheinol-derivates are common in Pluchea. More polar fractions of this extract contained Stigmast-5,22-dien-3-O-beta-D-glucopyranoside and the chlorophyll degradation products pheophorbide A and hydroxy-pheophorbide A. Previous publications identified some eudesmanolides as anti-inflammatory and anti neoplastic substances. Therefore, they may contribute to the cytotoxic effects of Pluchea odorata against various cancer cell lines.
