Die nukleäre Lamina erhält die mechanische Stabilität des Zellkerns in Metazoa. Ihre Hauptkomponenten sind die Lamine, die Teil der Intermediär-Filament-Protein-Familie sind. Da B-Typ Lamine permanent farnesyliert sind, konzentrieren sie sich ausschließlich an der Peripherie des Zellkerns. Hingegen liegen A-Typ Lamine auch als diffundierende Komplexe im Nukleoplasma vor, da ihre Farnesylierung durch posttranslationale Prozessierungen entfernt wird. Weiters, erfüllen Lamine Funktionen in vielen Prozessen im Zellkern. Daher ist es nachvollziehbar, dass Mutationen in Laminen, besonders in A-Typ Laminen, für zahlreiche Erbkrankheiten verantwortlich sind, wenn auch deren pathologische Mechanismen unbekannt sind. Lamine interagieren mit vielen anderen Proteinen, eines davon ist LAP2α, welches für den Erhalt der nukleoplasmatischen Lamin-Komplexe verantwortlich zu sein scheint. Die Funktion dieser Lamin-Komplexe ist unbekannt, jedoch sind mehrere Mutationen in Lamin A sowie eine in LAP2α mit dilatativer Kardiomyopathie assoziiert.
Bisher zeigten ausschließlich in vitro-Studien die Interaktion zwischen LAP2α und A-Typ Laminen. Deshalb, stelle ich hier ein für unterschiedliche Lamine adaptiertes bimolekulares Fluoreszenz-Komplementationssystem vor, sowie für mutante Lamine, welche mit Krankheiten assoziiert sind, und für LAP2α, mit dessen Hilfe Interaktionen zwischen diesen Komponenten in lebenden Zellen untersucht werden können. Es zeigte sich, dass alle untersuchten Lamine effizient miteinander interagieren. Die Bestätigung der Interaktion zwischen LAP2α und A-Typ Laminen gelang leider nicht, vermutlich aufgrund komplexer Interaktionen von Lamin-Komplexen.
Bisherige Immunofluoreszenz-Studien in fixierten Zellen und Geweben zeigten, dass, verglichen mit LAP2α-defizienten Zellen, in Zellen, welche LAP2α exprimieren, erhöhte Konzentrationen von nukleoplasmatischen A-Typ Laminen in der G1-Phase vorliegen, jedoch nicht in der S-Phase. Deshalb untersuchte ich die Dynamik EGFP-fusionierten Lamin As über den Zellzyklus in LAP2α-reduzierten Zellen und in Kontroll-Zellen. Dabei zeigte sich, dass LAP2α praktisch keinen Einfluss auf die Verteilung von Lamin A während der S- und der G2-Phase hatte. Daher und aufgrund weiterer Indizien ist es wahrscheinlich, dass LAP2α die Dynamik von Lamin A während der frühen G1-Phase beeinflusst.
Weiters, studierte ich die Dynamik neu-synthetisierter A-Typ Lamine. Die Expression von Konstrukten von pre-Lamin A und reifem Lamin A, zeigte, dass die transiente Farnesylierung von pre-Lamin A eine Rolle in dessen schneller Rekrutierung an die nukleäre Peripherie spielt, von wo ein Teil nach ungefähr zwei Stunden ins Nukleoplasma transloziert. Neu-synthetisiertes reifes Lamin A, welches nicht farnesyliert wird, reichert sich zunächst im Nukleoplasma an und wird dann langsam und kontinuierlich an die nukleäre Peripherie rekrutiert. Mutantes und Muskeldystrophie-assoziiertes pre-Lamin A-ΔK32 wurde bisher nie an der nukleären Peripherie beobachtet. In der vorliegenden Studie zeige ich jedoch, dass neu-synthetisiertes pre-Lamin A-ΔK32 tatsächlich aufgrund seiner C-terminalen Farnesylierung an die Peripherie rekrutiert wird, sich dann aber schnell ins Nukleoplasma verteilt.

The nuclear lamina maintains the structural integrity of the nucleus of the metazoan cell. Its main components are A- and B-type lamins, which are both members of the intermediate filament protein family. Unlike B-type lamins, which concentrate at the nuclear periphery due to their farnesylation at the C-terminus, A-type lamins are also found as diffusible complexes in the nucleoplasm as their farnesylation is removed during post-translational maturation. Moreover, lamins seem to play many roles in various nuclear processes. Therefore, it is understandable that mutations, mostly in A-type lamins, are responsible for a large variety of diseases, but their pathogenic mechanisms are hardly known. Lamins also interact with many other proteins. One of them is LAP2α, which seems to be important for the maintenance of the nucleoplasmic pool of A-type lamins. The function of this nucleoplasmic pool is largely unknown, however a mutation in LAP2α and some mutations in A-type lamins are associated with dilated cardiomyopathy.
To date, the interaction between LAP2α and A-type lamins was shown by in vitro-studies only. Therefore, I present here an adapted bimolecular fluorescence complementation system for various lamins and disease-linked mutants of lamins, and LAP2α, which enables the investigation of interactions between these components in living cells. I found that all studied lamins readily interacted with one another at the nuclear periphery. However, the binding of LAP2α to A-type lamins could not be shown with this approach probably due to the complex interplay between lamin-structures.
Furthermore, previous studies have shown by immunofluorescence microscopy in fixed cells and tissues that, compared to cells lacking LAP2α, cells expressing LAP2α showed increased levels of nucleoplasmic A-type lamins during G1-phase but not during S-phase. To investigate lamin-dynamics in living cells I monitored EGFP-tagged lamin A during the cell cycle in LAP2α-knockdown and control cells. I found that LAP2α had almost no effect on the distribution of lamin A during S- and G2-phase. Therefore, and due to additional indications, it is likely that LAP2α affects lamin-dynamics and localization in early G1-phase.
Moreover, I studied the dynamics of newly synthesized EGFP-tagged A-type lamins. Using constructs of pre-lamin A and mature lamin A showed that processing of prelamin A by transient farnesylation is involved in rapid targeting of newly synthesized lamins to the peripheral lamina, from where a fraction translocates to the nucleoplasm approximately two hours later. Newly synthesized mature lamin A, which is not farnesylated, accumulates first in the nucleoplasm and then steadily translocates to the nuclear lamina. The muscular dystrophy-associated mutant pre-lamin A-ΔK32 was previously never found to localize at the nuclear periphery. I present here that newly synthesized pre-lamin A-ΔK32 is in fact transiently recruited to the nuclear periphery via its C-terminal farnesylation, but subsequently distributes quickly into the nucleoplasm.
