Seit einigen Jahren werden Proteine gesucht die sich als Biomarker eignen, um Krankheiten, wie Krebs, in Blutproben nachweisen zu können. Ein Hauptproblem ist die geringe Konzentration potentieller Biomarker im Blut, verglichen mit den Hauptbestandteilen. Die Entwicklung noch leistungsfähiger analytischer Geräte ist nicht der einzige Weg niedrig konzentrierte Proteine analytisch zugänglich zu machen. Spezielle Probenvorbereitungstechniken sind notwendig um die Analyse niedrig konzentrierter Proteine zu unterstützen. Ein wesentlicher Faktor ist die limitierte Menge an Peptiden die in ein analytisches System, wie eine HPLC-MS, injiziert werden kann. Deshalb ist die Abtrennung oder zumindest Abreicherung der häufigsten Proteine im Blut ein vielversprechender Weg potentielle proteinogene Biomarker analytisch zugänglich zu machen. In dieser Arbeit werden Probenvorbereitungstechniken auf ihre Eignung hin untersucht den Nachweis und die Quantifizierung von niedrig konzentrierten Proteinen zu unterstützen. Anschließend werden die vielversprechendsten Techniken weiter optimiert. Die untersuchten Verfahren sind "Molecular-weight-cut-off"-Filter, SDS-PAGE und Depletion. Für jede der Techniken wurden mehrere Experimente durchgeführt. Die besten Resultate wurden bei einem der Depletionsversuche erzielt.

Since a couple of years proteins are searched, who may serve as biomarkers, to diagnose diseases, like cancer, within bloodplasma samples. A major problem is the low abundance of such possible biomarkers, compared to the main components of bloodplasma. The development of more powerful analytical instruments is not the only way to make low abundant proteins analyticaly accessible. Special sample preparation techniques are necessary to support the analysis of low abundant proteins. One crucial factor is the limited amount of total peptides that can be injected in the analytical system. So the separation or at least reduction of the most abundant proteins from the plasma sample is a promising way to allow the analysis of potential proteinogenic biomarkers. In this work different sample preparation techniques are investigated for their ability to support the detection and quantification of low abundant proteins. First some techniques are tested on there potential of supporting the detection of low abundant protein. Afterwards the most promising techniques where taken for further optimisation. The investigated techniques are molecular-weight-cut-off-filters, SDS-PAGE and depletion. For each of the techniques multiple experiments were done. The most promising results were received with depletion experiments.
