Das vorrangige Ziel dieser Arbeit war die funktionelle Charakterisierung des PCNA-Proteins, ein durch das Halovirus ϕCh1 kodiertes Protein. Die PCNA-Aminosäuresequenzen des ϕCh1 und seines Wirtes Natrialba magadii, ein halo- und alkaliphiles Archaeum, weisen einen hohen Ähnlichkeitsgrad auf, was auf einen horizontalen Gentransfer schließen lässt. Diese Untersuchungen haben gezeigt, dass das Hinzufügen von 21 Nukleotiden am N-Terminus unter der Kontrolle eines alternativen GTG Startcodons zu einer erhöhten Produktion an viralen Partikel und Proteinen sowie vermehrt zur Lyse der Kultur führt.(Diese in der Masterarbeit festgehaltenen Forschungsergebnisse wurden für die Erstellung eines Manuskriptes, welches auch inkludiert ist, verwendet.)
Darüber hinaus wurde eine heterozygote N. magadii L13 Mutante hergestellt, in welcher das Gen für einen Oberflächenrezeptor, der höchstwahrscheinlich eine Bindung des ϕCh1 ermöglicht, deletiert wurde. Des Weiteren wurde eine homozygotische Natronobacterium gregoryi Mutante erzeugt, bei der das Gen für NgAgo deletiert wurde. Außerdem wurde ein neuer E.coli Stamm, der haloalkaliphile Proteine exprimiert, kreiert. Zu guter Letzt wurden noch Stämme hergestellt, die für zukünftige Interaktionsstudien herangezogen werden können.

The primary focus of this thesis was to characterise the function of the PCNA protein encoded by the halovirus ϕCh1 which infects the haloalkaliphilic archaeon Natrialba magadii. The PCNA of ϕCh1 has a high amino acid sequence homology to the PCNA encoded by its host N. magadii indicating it has been acquired by horizontal gene transfer. Results indicate that the addition of a 21 nucleotide N-terminal tag, relative to the PCNA of N. magadii, under the control of an alternate GTG start codon significantly increases the production of viral proteins, the extent of culture lysis, and the production of viral particles. The results of this research have been used for the construction of a manuscript which is included in this thesis.
This thesis also includes several secondary investigations. These include the creation of a heterozygous deletion mutant of a potential binding target for ϕCh1 on the surface of N. magadii, the creation and analysis of a homozygous deletion mutant of the gene encoding NgAgo in Natronobacterium gregoryi, the redesign and construction of halophilic shuttle vectors, and the creation of a new strain of E. coli for the expression of haloalkaliphilic proteins and the construction of strains for the investigation of the interaction of the PCNA of ϕCh1 and the origin of replication of ϕCh1.
