In der vorliegenden Dissertation wurden die DNA-protektiven Effekte von Grüntee-Extrakt (GTE), Gallussäure (GA) und Vitamin C (VC), sowie verschiedener Kombinationen dieser Antioxidantien in inneren Organen von Ratten (Leber und Dickdarm) sowie in peripheren Blutzellen (Lymphozyten) in Einzelzellgelelektrophoresetests (SCGE Tests) untersucht. Die Substanzen wurden männlichen Ratten (n=5/Gruppe) in humanrelevanten Dosierungen über einen Zeitraum von fünf Tagen hinweg mittels Schlundsonde verabreicht (GTE 1,33mg/kg KG/T und 6,65mg/kg KG/T; GA 0,2mg/kg KG/T; VC 1,33mg/kg KG/T und 6,65mg/kg KG/T; Kombination aus 1,33mg GTE/kg KG/T, 0,2mg GA/kg KG/T und 6,65mg VC/kg KG/T). Die Verabreichung von 1,33mg GTE/kg KG/T (entspricht dem Konsum von einer Tasse Grüntee beim Menschen) löste keine Effekte aus, die fünffache Menge jedoch führte zu einer starken Abnahme der DNA-Schäden in den Blutzellen, in Leber- und Dickdarmzellen wurden schwächere Effekte gefunden. GA wirkte bereits in der niedrigen Dosierung (0,2mg/kg KG/T, ADI Wert beim Menschen) stark protektiv in Blutzellen und Dickdarmzellen, VC führte weder in der niedrigeren (DACH-Referenzwert) noch in der fünffach höheren Dosierung zu einer Abnahme der Schäden. Eine Kombination aller drei Antioxidantien (1,33mg GTE, 0,2mg GA und 6,65mg VC), löste ähnliche, aber schwächere Wirkungen als GA aus. Binäre Mischungen von GA und GTE nicht aber GA und VC reduzierten die DNA-Migration in Blutzellen und Dickdarmzellen. In einer weiteren Versuchsreihe wurden die Tiere nach Behandlung mit GA oder einer Kombination aller drei Antioxidantien bestrahlt (60Co Quelle, 30 Gy). Auch in diesem Experiment wurden DNA-protektive Wirkungen gefunden, und zwar nicht nur eine Abnahme der Bildung oxidierter Basen, sondern auch eine Reduktion der Einzel- und Doppelstrangbrüche. Zusätzlich wurden auch eine Reihe weiterer Parameter untersucht, und mit keinem der drei Antioxidantien wurde eine Änderung der LDL-Oxidation und der gesamten antioxidativen Kapazität (TEAC, gemessen mit ABTS) detektiert, auch immunologische relevante Endpunkte (TNF-α, IFN-γ, IL-4, IL-6, IL-10) wurden nicht beeinflusst. Diese Ergebnisse zeigen, dass GA in inneren Organen und auch in Blutzellen eine wesentlich stärker ausgeprägte Schutzwirkung vor oxidativen DNA-Schäden hat als VC und GTE.

Aim of the study was a comparison of the DNA-protective effects of green tea extract (GTE), gallic acid (GA) and vitamin C (VC) in inner organs (liver and colon) and in peripheral blood cells (lymphocytes) of rats in single cell gel electrophoresis (SCGE) assays.
The substances were given orally per gavage to male rats (n=5/group) over a period of five days in concentrations which are relevant for humans (GTE 1,3 mg/kg bw/d and 6,7 mg/kg bw/d; GA 0,2 mg/kg bw/d; VC 1,3 mg/kg bw/d and 6,7 mg/kg bw/d). Application of 1,3mg GTE/kg (according to consumption of one cup of green tea for humans) did not cause any effect, a fivefold concentration leads to a strong decrease of DNA-damage in blood cells. In liver and colon derived cells weak effects were found. GA (0,2mg/kg, ADI for humans) caused significant protection in blood and colon cells. VC did not induce a protective effect in the lower (DACH reference value for humans) or the fivefold dose. A combination of the three antioxidants (1,3 mg GTE, 0,2 mg GA and 3,7 mg VC) caused similar, but slightly less pronounced effects as compared to those seen with GA alone. A binary combination of GA (0,2 mg/kg) and GTE (1,3 mg/kg) reduced DNA damage in all organ investigated, whereas a combination of GA (0,2 mg/kg) and VC (6,7 mg) leads only to a marginal reduction of DNA migration only in liver cells.
In a second experiment, the animals were treated with GA and the combination of the three antioxidants and were subsequently γ-irradiatiated (60Co, 30 Gy). Also in this trial protective effects of GA and the combination were seen. The most pronounced effects were seen in the blood cells. It is notable that after radiation clear protective effects were also seen under standard SCGE-conditions (which reflect changes due to single- and double-strand breaks).
Additionally, a set of further parameters in the plasma of the rats was investigated, with none of the antioxidants, alterations of the LDL-oxidation and the total antioxidant capacity (TEAC, measured with the ABTS+ radical) were seen, also immunological parameters (TNF-α, IFN-γ, IL-4, IL-6 and IL-10) were not significantly altered.
Taken together, the findings of this study indicate that GA provides stronger protection against oxidative DNA damage in inner organs and blood cells than GTE and VC.
