Der marine Nematode Laxus oneistus und das sich in Längsrichtung teilende, stabförmige, gramnegative Bakterium Candidatus Thiosymbion oneisti sind die Hauptakteure einer rätselhaften Symbiose, die die Sandbänke im seichten Wasser rund um die Insel Carrie Bow Cay in Belize durchzieht. In einer vorrangegangen unveröffentlichte Studie konnte gezeigt werden, dass L. oneistus potenzielle antimikrobielle Proteine aus der Familie der Bactericidal Permeability Increasing Proteins (BPI) auf seiner Kutikula absondert und damit in Kontakt mit dem Symbionten kommt. Da beide Organismen nicht kultiviert werden können, ist es notwendig, die zu untersuchenden Proteine ektopisch zu exprimieren und ihre Funktion In vitro zu analysieren. Im ersten Teil dieser Studie haben wir rekombinante L. oneistus-BPIs in drei verschiedenen bakteriellen Expressionssystemen hergestellt und einen antimikrobiellen Test entwickelt, um die antimikrobielle Wirkung zu testen. Der zweite Teil dieser Arbeit konzentriert sich auf die Expression und funktionelle Charakterisierung von ParB, einem DNA-bindenden Protein, das von dem Symbionten hergestellt wird. ParB bindet die DNS-Sequenz parS und vermittelt die Chromosomentrennung in stäbchenförmigen Modellbakterien wie E. coli. In diesem Teil zeigen wir in einem Electrophoretic Mobility Shift Assay (EMSA), dass ParB an die parS Sequenz In vitro binden kann. Obwohl wir die antimikrobielle Aktivität der BPIs nicht nachweisen konnten, unterstützen unsere Daten die Rolle von ParB bei der Chromosomensegregation des Symbionten.
Dies ist der erste Bericht über ein in die Chromosomensegregation entlang der zellulären Längsachse involviertes ParB.

The nematode Laxus oneistus and the longitudinally dividing, rod-shaped gram-negative, Candidatus Thiosymbion oneisti are the main actors of a puzzling symbiosis pervading the shallow water sandbars around the island of Carrie Bow Cay, Belize. Previous unpublished work showed that L. oneistus secretes potential antimicrobial proteins belonging to the bactericidal permeability increasing protein (BPI) family onto its cuticle where it colocalizes with the symbiont. Since both organisms cannot be cultivated, it is imperative to express proteins recombinantly and study them in vitro in order to draw conclusions about their function. In the first part of this study, we produced recombinant L. oneistus BPIs in three different bacterial expression systems. Further, we developed an antimicrobial assay to test their antimicrobial effect.
The second part of this study focuses on the ectopic expression and functional characterization of ParB, a DNA-binding protein expressed by the symbiont. ParB binds parS sites and mediates chromosome segregation in model rod-shaped bacteria. Here, we produced recombinant ParB and, in an electrophoretic mobility shift assay, we showed that it binds the symbiont parS site in vitro.
Although we could not prove the antimicrobial activity of the nematode BPIs, our data support the role of symbiont ParB in chromosome segregation. This is the first report of this centromeric protein mediating chromosome segregation along the short cell axis of a bacterium.
