Mit weltweit ungefähr 650.000 Todesfällen durch Influenza und 1,6 Millionen Todesfällen durch Pneumokokken pro Jahr, stellt die Behandlung der respiratorischen Erkrankungen ausgelöst durch dieser beiden Erreger eine große Herausforderung dar (WHO, 2020a, 2020b).
In jüngsten Studien zeigten prenylierte Flavonoide aus der Maulbeerbaumwurzelrinde (Morus alba L.) eine hemmende Wirkung in vitro auf beide Krankheitserreger – Influenzavirus und Pneumokokken. Dies könnte in Zukunft eine wirksame Behandlungsstrategie darstellen. Um jedoch eine ausreichende Wirksamkeit und Sicherheit nachzuweisen, sowie Kenntnisse über Pharmakokinetik und Pharmakodynamik zu erhalten, sind in vivo Modelle erforderlich.
Ziel dieser Diplomarbeit war es daher, eine Methode zur Isolierung prenylierter Verbindungen aus der Maulbeerbaumwurzelrinde im Großmaßstab zu entwickeln, um die gewonnen Reinstoffe für in vivo Studien zur Verfügung zu stellen. Die Grundlage für diese Arbeit bildeten Fraktionen, die aus einer groben Auftrennung einer früheren Diplomarbeit bereits vorhanden waren. Unter Verwendung von Flash Chromatographie, Massenspektrometrie (MS) und nuklearen Magnetresonanz (NMR) Spektroskopie wurden vier Reinsubstanzen, namentlich Sanggenon C, D, G und Kuwanon L und eine semi-aufgereinigte Verbindung (Sanggenon O) in ausreichenden Mengen erfolgreich isoliert und strukturell identifiziert.

Respiratory infections due to influenza viruses and/or pneumococci are causing approximately 2.2 million deaths annually worldwide. Consequently, finding a treatment against these pathogens remains an ongoing challenge.
In recent studies, prenylated flavonoids, constituents of the white mulberry root bark (Morus alba L.), showed an in vitro inhibitory effect against influenza A virus as well as Streptococcus pneumoniae. This renders the plant material a highly promising starting point for the development of a herbal remedy. However, in order to prove sufficient effectiveness and safety, to obtain knowledge about pharmacokinetics and pharmacodynamics, in vivo models are required.
Therefore, the aim of this thesis was to develop a method for the large scale isolation of prenylated compounds of the mulberry root bark and to make them available for in vivo studies in sufficient amounts. This work is based on enriched fractions produced during an earlier diploma thesis. By means of flash chromatography, mass spectrometry (MS) and nuclear magnetic resonance (NMR) spectroscopy, four pure compounds, namely sanggenon C, D, G and kuwanon L and one semi-pure compound, i.e. sanggenon O, have been successfully isolated in sufficient amounts and structurally identified.
