Die globale Verschmutzung durch Plastikproduktion ist ein relevantes Thema der modernen Welt. Neben der Makroplastikverschmutzung stellen Miko- und Nanoplastikplastik (MNPs) eine noch nicht untersuchte Bedrohung für Tiere und Menschen dar. Über die Nahrung nehmen Menschen bis zu 5 g Plastik pro Woche zu sich. Die Aufnahme in den Körper und daraus resultierende gesundheitliche Risiken sind jedoch noch größtenteils unerforscht. Durch die Aufnahme über Nahrung ist das Verdauungssystem den höchsten Konzentrationen von MNPs ausgesetzt. Auswirkungen auf die Gesundheit wären also vermutlich am ehesten bei Darmerkrankungen zu erkennen. In dieser Arbeit wurde daher zuerst die Aufnahme von MNPs in Mäusen mit und ohne Colitis verglichen. Im Anschluss wurde der Einfluss von MNPs auf Darmkrebs untersucht. Für das Kurzzeit-MNP-Aufnahme-Experiment wurden wildtyp (wt) und DSS-behandelte (Colitis Model) C57BL/6J wt Mäuse verwendet. Nach einmaliger Zwangsfütterung von PS MNPs, wurden die Mäuse nach entsprechenden Zeitpunkten getötet und ihre Organe untersucht. APCmin/+ Mäuse wurden als Model für Darmkrebs verwendet. Die Überlebensdauer von Kohorten mit und ohne PS MNPs im Futter wurde verglichen. Nach ihrem Tod wurden die Organe entnommen und die Aufnahme in den Darm wurde untersucht. In allen analysierten Därmen der Mäuse des Colitis-Experiments wurden MNPs gefunden, wobei die mit DSS vorbehandelten Mäuse eine Tendenz zeigten, mehr und größere Partikel aufzunehmen. Nach 4h wurde am meisten Partikel im Darm entdeckt. Am häufigsten wurde Nanoplastik (d=0.293 µm) gefunden, vor Mikroplastik (d=1.14 µm). Größeres Mikroplastik (d= 10 µm) wurde nicht gefunden. In den APCmin/+ Mäusen senkte die Exposition mit MNPs die mittlere Überlebensdauer um rund 14 Tage, jedoch war dieser Trend nicht statistisch signifikant. Die Aufnahme der MNPs in das Darmgewebe war vergleichbar zu den Mäusen aus dem Colitis-Experiment, auch wenn keine Tendenz gefunden wurde, dass MNPs vermehrt von den Polypen aufgenommen werden. Die Auswirkungen auf die Bildung und den Schweregrad von Polypen werden zum Zeitpunkt dieser Arbeit noch genauer von Pathologen evaluiert. Zusammenfassend lässt sich sagen, dass PS MNPs von gesunden und kranken Mäusen in das Darmgewebe aufgenommen werden. Die genauen Mechanismen und Auswirkungen dort werden noch weiter erforscht.

The global pollution with plastic is a highly relevant topic. Next to macroplastic, micro- and nanoplastic (MNP) particles pose an uninvestigated threat to habitats, animals, and humans. As MNPs are inevitably contaminating the food chain, humans are estimated to ingest up to 5 g of plastic particles per week. The uptake, distribution and possible resulting health risks are, at the time of this thesis, mostly uninvestigated. The gastrointestinal tract (GIT) is the part of the body which is exposed to the highest MNP concentration. Hence, a possible effect on health would probably be seen in intestinal diseases. In this thesis, MNP uptake in mice with and without colitis was investigated and the influence of MNP on colorectal cancer (CRC) was examined. Wildtype (wt) and DSS pre-treated C57BL/6J mice (colitis model) were treated with a singule gavage of polystyrene (PS) MNPs and the timely uptake of PS MNPs into the GIT was examined. APCmin/+ mice were used as a model for CRC. The survival of a cohort with constant PS MNP treatment via food was compared to a control cohort. After being euthanized, organs were harvested and the PS MNP uptake into the GIT was examined. In the colitis experiment, MNPs were found in the GIT of all analyzed mice. DSS-treated mice showed a higher tendency to take up MNPs into the GIT (more particles and bigger particles). Overall, more nanoplastic (d=0.293 µm) was found then microplastic (d=1.14 µm). No microplastic of the size of 10 µm was found. Timewise, the highest amount of MNPs was found in the GIT of the mice sacrificed after 4h. This suggests an uptake of MNPs in the first 4h after the exposure, and an excretion or further distribution to other organs afterwards. The presence of polystyrene MNPs in the mouse diet seemed to lower the median overall survival of the APCmin/+ mice by about 14 days, however this trend was not statistically significant. Polystyrene MNPs were found in the SI and the Co tissue of all APCmin/+ mice in similar manner to the short-term experiment, though not specifically in polyps. Possible differences in the number and severity of polyp lesions are, as of summer 2023, currently under more detailed investigation by pathologists. In conclusion, polystyrene MNPs are shown to be taken up by the GI tissue, in both sick and healthy mice. The exact mechanisms, potential accumulation, and effects of MNP uptake are still under investigation.
