Die Synthese von organischen Phosphorverbindungen mit direkten Kohlenstoff-Phosphor (C-P) Bindungen sind in der chemischen Biologie von großem Interesse. Einerseits können sie dazu dienen die biologischen Abbauwege von Phosphonaten zu studieren und dabei helfen ein holistisches Bild des globalen Phosphorzyklus zu gewinnen. Andererseits sind organische Phosphorverbindungen, insbesondere in ihrer enantiomerenreinen Form, wichtige Bausteine von Wirkstoffen in der Medizin und Landwirtschaft. Deshalb wurden im Rahmen dieser Arbeit verschiedene Phosphon- und Phosphinsäuren hergestellt, die entweder als Substrate für enzym-mechanistische Studien oder als potenzielle Wirkstoffe von großer Bedeutung sind. Durch die Synthese von mono-, di- und trimethylierten Derivaten von 2-Aminoethylphosphonsäure und (R)-2-Amino-1-hydroxyethylphosphonsäure gelang es gemeinsam mit unseren Kooperationspartnern die Substrate der neu entdeckten Enzyme PbfB, PbfC und PbfD zu identifizieren und ihren Mechanismen aufzuklären. Außerdem wurde ein Set an Substanzen synthetisiert um C-P Lyase, den zwar am längsten bekannten, jedoch auch komplexeste Abbauweg für Phosphonate, zu untersuchen. Dazu wurde eine Phosphinsäure mit einem Fluorophor und einem passenden Quencher auf den beiden Seitenketten hergestellt, welche so bald eine der C-P Bindungen gespalten wird, eine einfache photometrische Methode zum Nachweis von C-P Lyase Aktivität darstellen könnte. Darüber hinaus wurde ein trifunktionelles Molekül, das eine Phosphonsäure-, eine Diazirin- und Alkingruppe enthält, synthetisiert, welches als alleinige Phosphorquelle für E. coli, die C-P Lyase exprimieren, dienen kann. Durch Aktivierung mittels UV-Licht in vitro gelang es, die Substanz kovalent an die beteiligten Proteine zu binden und diese nach Aufreinigung entweder mit einem Fluorophor zu koppeln oder direkt mittels LC-MS/MS zu untersuchen. Bezüglich der Synthese pharmazeutisch- oder landwirtschaftlich relevanter Phosphonsäuren gelang es, den kürzlich entdeckten Naturstoff Pantaphos in racemischer Form herzustellen. Diese Verbindung besitzt ein großes Potential als Herbizid, aber seine genaue dreidimensionale Struktur ist noch unbekannt. Deshalb wurde eine Synthesemethode entwickelt, die in Zukunft die Synthese beider Enantiomere von Pantaphos ermöglichen wird, um sie für biologische Studien zugänglich zu machen. Des Weiteren wurde eine allgemeine Methode zur Synthese der meisten Aminophosphonsäureanaloga zu den proteinogenen Aminocarbonsäuren in sehr hoher Enantiomerenreinheit entwickelt, die auf einer asymmetrischen Transferhydrierung als Schlüsselschritt zur Herstellung hoch enantiomerenangereicherter α-Hydroxyphosphonate beruht. Neben der Synthese medizinisch relevanter Moleküle erlaubt diese Methode auch die stereoselektive Deuterierung in α-Position zum Phosphor. Dadurch erleichtert die entwickelte Methodik mechanistische Studien an Enzymen aus dem Phosphonatmetabolismus erheblich.

Synthetic samples of organic phosphorus compounds with direct carbon-phosphorus (C-P) bonds are of high interest in chemical-biology research. On the one hand, they are needed to gain knowledge about microbial degradation pathways of phosphonates, this is crucial to understand the global phosphorus cycle. On the other hand, organophosphorus compounds, especially in their enantiomerically pure form, are important building blocks in medicine and agriculture. Therefore, within the framework of this thesis, various phosphonic and phosphinic acids were prepared as probes for enzyme mechanistic studies or as potential future drug candidates. By synthesizing mono-, di-, and trimethylated derivatives of 2-aminoethylphosphonic acid and (R)-2-amino-1-hydroxyethylphosphonic acid, the substrates, and the mode of action of the newly discovered enzymes PbfB, PbfC and PbfD was unraveled. Additionally, a set of substances was synthesized to study C-P lyase, the longest known but also most complex phosphonate degradation pathway. These include a phosphinic acid containing a fluorophore and a suitable quencher on separate side chains which, once one of the C-P bonds is cleaved, will presumably provide a simple photometric method to detect C-P lyase activity in complex biological samples. Furthermore, a trifunctional compound was synthesized bearing a phosphonic acid, a diazirine and an alkyne moiety, which can serve as sole phosphorus source for E. coli by action of C-P lyase. By activation with UV-light in vitro, it was possible to covalently bind this substance to the involved proteins, which were either coupled with a fluorophore or used for LC-MS/MS studies after purification. Regarding phosphonic acids with a potential application spectrum in medicine and agriculture, the recently discovered biogenic molecule pantaphos was successfully synthesized as a racemate. This compound has great potential as herbicide, but its exact three-dimensional structure is yet unknown. Thus, a synthetic method was developed that will in the future allow for the synthesis of both enantiomers of pantaphos for biological studies. In addition, a general method for the synthesis of most aminophosphonic acid analogs of the proteinogenic aminocarboxylic acids in very high enantiomeric purity was developed by relying on asymmetric transfer hydrogenation as the key step to produce highly enantioenriched α-hydroxyphosphonates. Beside the synthesis of medically relevant molecules, the developed method also allows for the stereoselective deuteration in α-position to the phosphorus. This has the potential to greatly facilitate mechanistic studies on phosphonate metabolizing enzymes.
