α-Aminophosphonsäuren und α-Aminophosphonate sind Verbindungen, die als Arzneimittel von großem Interesse sind. Daher ist es von größter Bedeutung, neue und innovative Synthesewege zu finden, um diese Verbindungen in ihrer enantiomerenreinen Form zu erhalten. Die meisten in der Literatur bekannten Synthesemethoden decken jedoch nur ein begrenztes Substratspektrum ab, verwenden gefährliche Chemikalien oder greifen auf teure chirale Poolreagenzien zurück. Daher konzentriert sich diese Masterarbeit auf die Entwicklung einer einfachen, katalytischen Methode, die die Synthese eines breiten Spektrums von α-Aminophosphonsäuren ermöglicht und eine Vielzahl von funktionellen Gruppen toleriert. Die Arbeit ist in drei Hauptteile gegliedert. Der erste Teil beschäftigt sich mit der Synthese von enantiomerenreinem (R)-Phosphalysin [(R)-91] aus einem einfachen Ausgangsmaterial, 4-Pentensäure (85). 85 wird über eine Arbuzov-Reaktion zu Diisopropyl-pent-4-enoylphosphonat (86) umgesetzt, das in einer asymmetrischen Transferhydrierung mit Noyori-Katalysator (R,R)-RuCl[(p-Cymol)-TsDPEN] [(R,R)-97] zu chiralem (S)-Diisopropyl-(1-hydroxy-4-pentenyl)-phosphonat [(S)-87] reduziert werden kann. Dies führt Chiralität in das Gerüst ein und ist der Schlüsselschritt des Syntheseweges. (S)-87 durchläuft eine Hydroborierungs-/Oxidationsreaktion, bei der eine endständige Hydroxylgruppe am Gerüst installiert wird, wodurch (S)-Diisopropyl-(1,5-dihydroxypentyl)phosphonat [(S)-88] entsteht. Die beiden Hydroxylgruppen können in einer Mitsunobu-Reaktion mit HN3 durch Azidogruppen ersetzt werden. Diese Reaktion führt auch die gewünschte (R)-Konfiguration am α-Kohlenstoff neben dem Phosphor ein. Die anschließende Hydrierung mit H2 und Pd/C und die Entschützung der Phosphonatgruppen ergibt (R)-91. Der zweite Teil der Arbeit befasst sich mit der Untersuchung alternativer Nukleophile, die anstelle des giftigen und explosiven HN3 in der durchgeführten Mitsunobu-Reaktion verwendet werden könnten. Zu diesem Zweck wurden Maleimid, Trimethylsilylazid, Phthalimid und Tetrachlorphthalimid untersucht. Schließlich erwies sich Tetrachlorphthalimid als der vielversprechendste Kandidat. Auf diese Weise wurde (R)-Diisopropyl-(1,5-bis(4,5,6,7-tetrachloro-1,3-dioxoisoindolin-2-yl)pentyl)phosphonat [(R)-112], ein möglicher Vorläufer von (R)-91, synthetisiert. Der dritte Teil der Arbeit befasst sich mit der Synthese der (R)-Phosphaarginin-Vorstufen (R)-Diisopropyl-4-hydroxy-1-((S)-4,5,6,7-tetrachloro-1-hydroxy-3-oxoisoindolin-2-yl)butyl)phosphonat [(R, S)-113] und (R)-Diisopropyl-4-hydroxy-1-((R)-4,5,6,7-tetrachlor-1-hydroxy-3-oxoisoindolin-2-yl)butyl)phosphonat [(R,R)-113] und baut auf den in den Teilen eins und zwei erworbenen Kenntnissen auf. Dieser Syntheseweg beginnt mit 85 und teilt seine ersten drei Schritte mit der Synthese von (R)-91. Das Schlüsselzwischenprodukt (S)-87 wird in einer Mitsunobu-Reaktion mit Tetrachlorphthalimid umgesetzt. Das Produkt dieser Reaktion wird dann in einer Ozonolyse mit reduktiver Aufarbeitung modifiziert, um das C5-Backbone um ein C-Atom zu verkürzen und gleichzeitig eine endständige Hydroxylgruppe einzubauen, wodurch die Vorstufen (R,S)- und (R,R)-113 entstehen.

α-Aminophosphonic acids and α-aminophosphonates are compounds of high interest as drugs. Hence, it is of utmost importance to find new and innovative synthetic pathways to obtain these compounds in their enantiopure form. However, most literature known methods for their synthesis only cover limited substrate scopes, use hazardous chemicals or make use of expensive chiral pool reagents. Thus, this master thesis focuses on the development of a simple, catalytic method that allows for the synthesis of a broad scope of α-aminophosphonic acids and tolerates a variety of functional groups. The thesis has three main parts. The first part concerns itself with the synthesis of enantiopure (R)-phosphalysine [(R)-91] from a simple starting material, 4-pentenoic acid (85). 85 is converted via an Arbuzov reaction to diisopropyl pent-4-enoylphosphonate (86), which can be reduced in an asymmetric transfer hydrogenation with Noyori-catalyst (R,R)-RuCl[(p-cymene)-TsDPEN] [(R,R)-97] to chiral (S)-diisopropyl (1-hydroxy-4-pentenyl)phosphonate [(S)-87]. This introduces chirality into the scaffold and is the key step of the synthetic pathway. (S)-87 undergoes a hydroboration/oxidation reaction, which installs a terminal hydroxyl-group on the scaffold, affording (S)-diisopropyl (1,5-dihydroxypentyl)phosphonate [(S)-88]. The two hydroxyl-groups can be replaced by azido-groups in a Mitsunobu reaction with HN3. This reaction also introduces the desired (R)-configuration on the α-carbon next to phosphorus. Subsequent hydrogenation with H2 and Pd/C and deprotection of the phosphonate groups affords (R)-91. The second part of the thesis deals with an investigation of alternative nucleophiles that could be used instead of the toxic and explosive HN3 in the performed Mitsunobu reaction. Maleimide, trimethylsilyl azide, phthalimide and tetrachlorophthalimide were investigated for this purpose. Finally, tetrachlorophthalimide turned out to be the most promising candidate. (R)-Diisopropyl (1,5-bis(4,5,6,7-tetrachloro-1,3-dioxoisoindolin-2-yl)pentyl)phosphonate [(R)-112], a possible precursor to (R)-91 was synthesized this way. Part three of the thesis deals with the synthesis of (R)-phosphaarginine precursors (R)-diisopropyl 4-hydroxy-1-((S)-4,5,6,7-tetrachloro-1-hydroxy-3-oxoisoindolin-2-yl)butyl)phosphonate [(R,S)-113] and (R)-diisopropyl 4-hydroxy-1-((R)-4,5,6,7-tetrachloro-1-hydroxy-3-oxoisoindolin-2-yl)butyl)phosphonate [(R,R)-113] and it builds upon knowledge acquired in parts one and two. This synthetic pathway starts with 85 and shares its first three steps with the synthesis of (R)-91. Key intermediate (S)-87 is reacted with tetrachlorophthalimide in a Mitsunobu reaction. The product of this reaction is then modified in an ozonolysis with reductive work-up to shorten the C5-backbone by one C-atom and install a terminal hydroxyl-group at the same time, affording precursors (R,S)- and (R,R)-113.
