Die Resistenz gegen Daptomycin, ein wichtiges Antibiotikum für die Behandlung grampositiver Staphylococcus aureus Infektionen, ist ein wachsendes Problem in der klinischen Praxis. Obwohl DAP als Antibiotikum der letzten Wahl gilt, ist dennoch eine Resistenz entstanden. Es wird angenommen, dass die Lipidzusammensetzung der Zellmembran von Staphylokokken eine Rolle bei ihrer Fähigkeit spielt, gegen Daptomycin resistent zu sein. In diesem Projekt wurde daher die Rolle der Lipide Phosphatidylglycerol, Lysyl-Phosphatidylglycerol und insbesondere Cardiolipin, die wichtigen Bestandteile der Membran von Staphylococcus aureus sind, bei der Daptomycin-Resistenz untersucht. Mit Hilfe biophysikalischer Methoden wie Langmuir-Trog-Monolayer-Kompression, Isothermal Titration Calorimetry und Zeta-Potential-Analysen wurden die Wechselwirkungen und strukturellen Eigenschaften verschiedener Mischungen dieser Lipide untersucht. Interessanterweise zeigten die Ergebnisse, dass Cardiolipin einen wichtigen Einfluss auf die Flexibilität und strukturelle Komplexität der Monoschicht hat, obwohl es nur in geringen Mengen in der Membran vorhanden ist. Dennoch wurde festgestellt, dass Cardiolipin keinen signifikanten Einfluss auf die Bindungsaffinität von Daptomycin hat. Die Analyse der Druck-Flächen-Isotherme der Monoschicht deutet darauf hin, dass Cardiolipin eine negative Krümmung begünstigt, aber eine ebene Monoschicht-Grenzfläche in Gegenwart von LPG toleriert. Diese Ergebnisse geben Einblicke in die Rolle spezifischer Lipide bei den molekularen Mechanismen der Daptomycin-Resistenz und unterstreichen die komplexe Rolle der Lipide bei der Antibiotikawirkung.

Resistance to daptomycin, an important antibiotic for the treatment of Gram-positive Staphylococcus aureus infections, is a growing problem in clinical practice. Although DAP is considered an antibiotic of last resort, resistance has nevertheless emerged. The lipid composition of the cell membrane in staphylococci, is thought to play a role in their ability to resist daptomycin. This project therefore investigated the role of the lipids posphatidylglycerol, lysyl-phosphatidylglycerol and in particular cardiolipin, which are important components of the Staphylococcus aureus membrane, in daptomycin resistance. By using biophysical methods such as Langmuir-trough monolayer compression, Isothermal Titration Calorimetry and Zeta potential analyses, the interactions and structural properties of different mixtures of these lipids were investigated. Interestingly, the results showed that cardiolipin exhibits an important influence on monolayer flexibility and structural complexity despite being present in low quantities in the membrane. Nevertheless, cardiolipin was found to have no significant effect on the binding affinity of daptomycin. Monolayer pressure-area isotherm analysis suggested that cardiolipin facilitates negative curvature but tolerates a planar monolayer interface in the presence of LPG. These results provide insights into the roles of specific lipids on the molecular mechanisms of daptomycin resistance and emphasize the complex role of lipids in antibiotic action.
