Donepezil und Rivastigmin sind zwei bedeutsame cholinerge Therapeutika, die primär zur Linderung der kognitiven Symptome bei Alzheimer und anderen Demenzformen zugelassen sind. Die Notwendigkeit neuer Alzheimer-Medikamente ergibt sich aus der begrenzten Halbwertszeit und den auftretenden Nebenwirkungen. Für diese Studie wurden 26 neue und vielversprechende Donepezil und Rivastigmin Hybride auf deren in vitro Plasmastabilität in Human- und Rattenplasma untersucht. Dabei wurden die Plasmaproben über einen Zeitraum von 24.00 h bei einer Temperatur von 37 °C inkubiert und mit UHPLC oder LC-HRMS vermessen. Wie erwartet, zeigten die meisten Analyten, sowie AN13 (89.5±1.4% in Humanplasma and 75.3±9.4% in Rattenplasma), einen stärkeren Abbau in Rattenplasma als in Humanplasma nach 24.00 h Inkubation. Die Derivate GIA25, ROS111 und VIT35 zeigten ein gegenteiliges Verhalten. Nach 24.00 h Inkubation verblieben 67.9±6.5% von GIA25 in Humanplasma, verglichen mit den 74.7±4.1% in Rattenplasma. Ähnlich blieben 84.3±2.6% von ROS111 und 67.9±6.5% von VIT35 im Humanplasma, während im Rattenplasma 89.6±3.4% und 74.7±4.1% erhalten geblieben sind. Der Analyt ROS131 zeigte die kürzeste berechnete in vitro t1⁄2, mit über 24.00 h im Humanplasma (circa 39 h) und 17 h im Rattenplasma. Grundsätzlich haben die Derivate eine größere Plasmastabilität als der Parentcompound Rivastigmine (t1⁄2 = 1 h) gezeigt. Zwanzig Analyten übertrafen mit ihrer berechneten t1⁄2 Humanplasma in vitro die in vivo t1⁄2 von Donepezil (t1⁄2 = 80 h) im Menschen, darunter AN13 (t1⁄2 = 173 h), AN15 (t1⁄2 = 161 h) und ROS151 (t1⁄2 = 169 h). Die Analyten haben in dieser Stabilitätsassay den Parentcompound Rivastigmin durchwegs übertroffen, in Hinblick auf die t1⁄2. In den meisten Fällen wiesen die Derivate eine höhere t1⁄2 im Humanplasma als der Parentcompound Donepezil.

Donepezil and rivastigmine are two cholinergic therapeutics that are approved for the treatment of cognitive symptoms associated with Alzheimer's disease and other forms of dementia. The need for optimised Alzheimer's drugs arises from the limited half-life and the adverse effects that occur. In this study, novel and promising 26 donepezil and rivastigmine hybrids were analysed for their in vitro plasma stability in human and rat plasma. The plasma samples were incubated at a temperature of 37 °C for a period of 24.00 h and measured using UHPLC or LC-HRMS. As expected, most of the analytes including AN13 (89.5±1.4% in human plasma and 75.3±9.4% in rat plasma) degraded faster in rat plasma than in human plasma following the 24.00 h of incubation. The derivatives GIA25, ROS111 and VIT35 showed the opposite trend. After 24.00 h incubation 67.9±6.5% of GIA25 remained in human plasma compared to 74.7±4.1% in rat plasma. Similarly, 84.3±2.6% of ROS111 and 67.9±6.5% of VIT35 remained in human plasma, compared to 89.6±3.4% and 74.7±4.1%, respectively, in rat plasma. The analyte ROS131 had the shortest calculated in vitro t1⁄2 with more than 24.00 h in human (approximately 39 h) and 17 h in rat plasma. Overall, the derivatives demonstrated higher plasma stability than the parent compound rivastigmine (t1⁄2 = 1 h) . Twenty analytes surpassed the human in vivo t1⁄2 of donepezil (t1⁄2 = 80 h) with their calculated t1⁄2 in the human plasma in vitro study including AN13 (t1⁄2 = 173 h), AN15 (t1⁄2 = 161 h) and ROS151 (t1⁄2 = 169 h). The compounds consistently outperformed the parent compound rivastigmine in this stability assay in terms of t1⁄2. In most cases, the derivatives displayed a superior t1⁄2 in human plasma than the parent compound donepezil.
