Die neuromuskuläre Synapse (NMS) stellt die Verbindungsstelle zwischen einem Motorneuron und einer Muskelfaser dar. Agrin, ein von Motorneuronen sezerniertes Protein, der Agrin-Rezeptor Lrp4 und die Muskel-spezifische Kinase (MuSK) nehmen eine zentrale Rolle bei der Entstehung der NMS ein. Agrin aktiviert über Lrp4 und MuSK eine Signalkaskade, die zur Aggregation von Proteinen einschließlich Acetylcholin-Rezeptoren (AChR) auf der postsynaptischen Seite der NMS führt. Ein bis dato unbekanntes Protein, vorläufig Cl-6 genannt, wurde als MuSK-Bindungspartner identifiziert. Eine Sequenzanalyse zeigte, dass dieses Protein homolog zu den Proteinen der RIN Familie ist. Aufgrund dieser Homologie und der zellulären Lokalisierung von Cl-6, nehmen wir an, dass Cl-6 eine regulatorische Rolle in der Endozytose von MuSK hat. Im Rahmen dieser Diplomarbeit wurde die Interaktion von Cl-6 mit MuSK and Rab5 untersucht and die verantwortlichen Regionen mit Hilfe von „GST pull-downs“ identifiziert. Die Vps9 Domäne wurde als Interaktionsdomäne für Rab5 gefunden, was drauf hinweist, dass Cl-6 vielleicht auch eine Guanin Nukleotid Austauschfaktor Aktivität wie die anderen RIN Proteine, besitzt. Zusätzlich wurden stabile Muskelzelllinien, die verschiedene RNA Interferenz Konstrukte trugen, verwendet um zu zeigen, dass die spezifische Herunterregulierung von Cl-6 zu einer verminderten Aggregation der AChR führt. Diese Ergebnisse führen zu der Schlussfolgerung, dass Cl-6 eine wichtige nachgeordnete Komponente im Agrin/Lrp4/MuSK Signalweg ist, die einen Einfluss auf die Aggregation der AChR an der NMS hat.

The neuromuscular synapse (NMS) represents the contact point between a motor neuron and a muscle fiber. The formation of the NMS depends on Agrin, a protein secreted by motor neurons, Lrp4, which is the Agrin-receptor, and MuSK (muscle-specific kinase). Agrin activates through Lrp4 and MuSK a signaling cascade, which leads to the aggregation of proteins, including acetylcholine receptors (AChRs), at the postsynaptic site of the NMS. A so far unknown protein, currently termed Cl-6, was found as an interaction partner of MuSK. Sequence analysis revealed that this protein is homologous to the proteins of the RIN family. Due to this homology and the cellular localization of Cl-6, we hypothesize that Cl-6 plays a regulatory role in MuSK endocytosis. In the context of this diploma thesis the interaction of Cl-6 with MuSK and Rab5 has been investigated and the responsible binding region was narrowed down using a GST pull-down assay. The Vps9 domain was identified as the binding region between Cl-6 and Rab5, suggesting that Cl-6 might also possess guanine nucleotide exchange factor activity, like the other RIN proteins. Additionally, stable muscle cell lines carrying different RNA interference constructs were used to show that a specific downregulation of Cl-6 results in reduced clustering of AChRs. This results lead to the conclusion that Cl-6 represents an important component in the downstream signaling of the Agrin/Lrp4/MuSK signaling pathway, which influences the clustering of AChRs at the NMS.
