Origanum vulgare und O. majorana sind zwei der bekanntesten Arten der Gattung Origanum. Das Enzym γ-Terpinensynthase kommt in beiden Arten vor und ist maßgeblich an der Produktion der Monoterpene Thymol und Carvacrol, den Hauptbestandteilen des Ätherischen Öls der Cymyl-Chemotypen von O. vulgare beteiligt. Das Ätherische Öl von O. majorana enthält kein Carvacrol und wird dem Sabinyl-Typ mit cis-Sabinenhydrat als geschmacksgebende Komponente zugeordnet. Im Zuge dieser Arbeit wurden die γ-Terpinensynthasen von O. vulgare und O. majorana untersucht, um herauszufinden, ob Thymol und Carvacrol in O. majorana aufgrund einer nicht funktionalen γ-Terpinensynthase nicht produziert werden kann. Es wurden 68 Klone des γ-Terpinensynthase Gens, 20 aus O. vulgare und 48 aus O. majorana, isoliert und sequenziert, wobei Unterschiede in Länge und Basenabfolge der DNA-Sequenzen gefunden wurden. Die innerartliche Variabilität der Klone aus O. vulgare ist größer als die der Klone aus O. majorana. Es konnte keine offensichtliche Ursache, wie z.B. Mutationen, die zu internen Stopp-Codons führen, für die unterschiedliche Monoterpenproduktion in O. vulgare und O. majorana gefunden werden, da in beiden Arten das TPS2-Gen intakt scheint. Interne Stopp-Codons waren zwar in beiden Arten vorhanden, jedoch waren die scheinbar intakten, vollständigen Sequenzen deutlich in der Überzahl. Mit verschiedener Software wurden hohe Wahrscheinlichkeiten für das Vorhandensein und Funktionieren des Transportproteins für den Membrantransport in O. majorana berechnet. Auffällige Mutationen in Exon 1, die das Transportprotein beeinträchtigen könnten, konnten bisher nicht als Ursache identifiziert werden. Die zahlreichen Mutationen in Intron 1, die in die Genregulation eingreifen könnten, wären ebenfalls ein möglicher Ausgangspunkt für weiterführende Untersuchungen.

Origanum vulgare and O. majorana are two of the best-known species of the genus Origanum. The enzyme γ-terpinene synthase is found in both species and is significantly involved in the production of the monoterpenes thymol and carvacrol, the main components of the essential oil of the cymyl-chemotypes of O. vulgare. O. majorana, however, produces no cymyl-compounds but is dominated by Sabinyl-compounds, sensorially important for the typical aroma of marjoram. In this work, the γ-terpinene synthases of O. vulgare and O. majorana were investigated to find out whether thymol and carvacrol are not produced in O. majorana due to a nonfunctional γ-terpinene synthase. 68 clones of the γ-terpinene synthase gene, 20 from O. vulgare and 48 from O. majorana, were isolated and sequenced, revealing differences in the length and base order of the DNA sequences. The intraspecific variability of the clones from O. vulgare is greater than that of the clones from O. majorana. No evident reason, such as mutations leading to internal stop codons, could be found for the different monoterpene production in O. vulgare and O. majorana, as the TPS2 gene appears intact in both species. Internal stop codons were present in both species, but the apparently intact, complete sequences were in majority. Specific software calculated high probabilities for the presence and functionality of the transport protein for membrane transport in O. majorana. Conspicuous mutations in exon 1, that could affect the transport protein, have not yet been identified as a cause. The numerous mutations in intron 1, that could interfere with gene regulation, would also be a possible starting point for further investigations.
