Die neotropische Gattung Notopleura (Benth. & Hook. f.) Bremek (Rubiaceae), die etwa 100 Arten ledrige bis sukkulente Kräuter und Sträucher umfasst, ist ein interessanter Gegenstand für phytochemische Untersuchungen. Zahlreiche Arten der Rubiaceae-Familie, darunter auch Arten der Gattung Notopleura, haben aufgrund ihrer komplexen biochemischen Stoffwechselwege und einer breiten Palette an speziellen Pflanzenmetaboliten in einer Vielzahl von Kulturen eine wichtige ethnomedizinische Bedeutung. Diese reiche Vielfalt an Metaboliten hat die phytochemische Forschung innerhalb der Familie seit langem vorangetrieben. Doch obwohl die Rubiaceae sich als vielversprechende Quelle für bioaktive Verbindungen erwiesen haben, wurde die Gattung Notopleura bisher noch nicht umfassend aus einem breit gefächerten, vergleichenden phytochemischen Blickwinkel untersucht. Die vorliegende Studie versucht, diese Lücke zu schließen, indem sie die in Notopleura-Arten gefundenen speziellen Pflanzenmetaboliten untersucht und ihre mögliche Bedeutung bewertet. Anhand von chromatographischen Analysen unter Anwendung von Hochleistungsflüssigkeits-chromatographie (HPLC) und des Gesamtphenolgehalts, charakterisiert die Studie die chemischen Profile von 16 verschiedenen Notopleura-Arten und vergleicht diese weiters mit analytischen Daten von Arten aus fünf nahe verwandten Gattungen. Diese vergleichende Analyse ergab eine chemische Unterscheidung zwischen den beiden Untergattungen von Notopleura. Darüber hinaus wurden die vier Substanzen Harounosid (1), 2-Azaanthrachinon (2), das Anthrachinonderivat 6-Hydroxy-2-aza-anthrachinon (3) und das Neolignanderivat Glochidiobioside (4) erfolgreich in N. panamensis identifiziert. Glochidiobioside (4) wurde erstmals in einer Notopleura Art nachgewiesen und das 6-Hydroxy-2-aza-anthrachinon (3), ist bisher noch nicht als Naturstoff beschrieben. Dieses Ergebnis unterstreicht das Potenzial von Notopleura als Quelle einzigartiger spezieller Pflanzenstoffe. Darüber hinaus wurden Fraßhemmungs-Tests mit polyphagen Raupen durchgeführt, um die ökologische Relevanz der beiden zuvor identifizierten Verbindungen Harounoside (1) und 2-Aza-anthrachinon (2) zu bewerten und Einblicke in ihre mögliche Rolle bei der Pflanzenabwehr zu gewinnen. Diese Ergebnisse machen deutlich, dass weitere Studien erforderlich sind, um die pharmakologische und ökologische Bedeutung der speziellen Metaboliten von Notopleura umfassend zu klären und unser Wissen über die chemische Vielfalt innerhalb der Rubiaceae erheblich zu erweitern.

The neotropical genus Notopleura (Benth. & Hook. f.) Bremek (Rubiaceae), comprising approximately 100 species of leathery to succulent herbs and shrubs, represents an interesting subject for phytochemical investigation. Numerous species in the Rubiaceae family, including species from the genus Notopleura, have an important ethnomedical relevance in a variety of cultures due to their complex biochemical pathways and wide range of specialized metabolites. This rich metabolite diversity has long fueled phytochemical research within the family. But even though Rubiaceae has shown promise as a source of bioactive compounds, the genus Notopleura has not yet been thoroughly studied from a wide-ranging, comparative phytochemical point of view. By examining the specialized metabolites found in Notopleura species and assessing their possible importance, this study seeks to close this gap. Based on chromatographic analyses using high-performance liquid chromatography (HPLC) and the determination of the total phenolic content, the study characterizes the chemical profiles of 16 different Notopleura species, comparing them with analytical data of five closely related genera. This comparative analysis revealed a chemical distinction between the two subgenera of Notopleura. Furthermore, the four substances harounoside (1), 2-aza-anthraquinone (2), its derivative 6-hydroxy-2-aza-anthraquinone (3) and the neolignane derivative glochidiobioside (4) were successfully identified from N. panamensis. Glochidiobioside (4) represents a novel discovery within the genus, and 6-hydroxy-2-aza-anthraquinone (3) is currently an undescribed compound. These findings highlight the potential of Notopleura as a source of unique specialized metabolites. Additionally, antifeedant assays employing polyphagous caterpillars were conducted to assess the ecological relevance of the two previously identified compounds harounoside (1) and 2-aza-anthraquinone (2), providing insights into their potential role in plant defense. These results highlight the need for more studies to clarify the pharmacological and ecological significance of Notopleura's specialized metabolites and significantly broaden our knowledge of the chemical diversity within Rubiaceae.
