Die räumliche Trennung von Transkription und Translation ist eines der charakteristischen Merkmale von Eukaryoten, das durch den Zellkern ermöglicht wird. In den letzten Jahren wurden jedoch virale Strukturen beschrieben, die funktionelle und strukturelle Ähnlichkeiten mit dem eukaryotischen Zellkern aufweisen, und für die eine räumliche Trennung von Transkription und Translation postuliert wurde. Viele Riesenviren replizieren im Zytoplasma der Wirtzelle in sogenannten viral factories. Die Architektur und räumliche Organisation vieler Replikationsschritte innerhalb dieser viral factories ist bis heute unklar. Es wurde gezeigt, dass die Mimivirus viral factory aus zwei biomolekularen Kondensaten besteht, die als äußere und innere Schicht bezeichnet werden. In dieser Arbeit untersuchen wir die Entwicklung der äußeren Schicht im Laufe der Infektion, um die Architektur der viral factory und potenzielle Rollen dieses Kondensats besser zu verstehen. Wir haben ein Protokoll zur single molecule in situ hybridization (smFISH) entwickelt, das spezifisch virale messenger RNA (mRNA)markiert und es uns ermöglicht mRNA-Lokalisierung in späten Infektionsstadien sichtbar zu machen. In Kombination mit bio-orthoginal amino acid tagging und FISH an ribosomaler RNA der Wirtzelle sind wir in der Lage Transkription und Translation visuell zu unterscheiden. Mit diesen Methoden zeigen wir eine räumliche Trennung von Transkription und Translation. Wir liefern außerdem erste Hinweise, dass die äußere Schicht in diesen Prozessen involviert ist. Diese Arbeit liefert neue Werkzeuge für die Erforschung von Riesenvirusinfektionen sowie Einblicke in die Architektur und Abläufe im inneren der viral factory.

Spatial separation of transcription and translation is one of the hallmark features of eukaryotes, facilitated by the nucleus. In recent years, however, viral structures were described that appear to have structural and functional similarities to the nucleus, and a spatial separation of transcription and translation has been proposed. Many giant viruses replicate in the cytoplasm in so-called viral factories. The architecture and spatial organization of many replication steps remain obscure to this day. The Mimivirus factory has been shown to be made up of two biomolecular condensates called the outer and inner layers. In this thesis, we investigate the progression of the outer layer throughout the infection to better understand the architecture of the viral factory and the potential role of this condensate. We have developed a single-molecule in situ hybridization protocol to specifically target Mimivirus messenger RNA (mRNA), allowing us to visualize mRNA localization in late-stage infections. Combined with bio-orthogonal amino acid tagging and ribosomal RNA fluorescence in situ hybridization we were able to distinguish between transcription and translation. We thereby show spatial separation of transcription and translation and provide preliminary evidence linking the outer layer condensate to these processes. This thesis provides novel tools for studying giant virus infections in the future as well as insights into the architecture and inner workings of the viral factory.
