In der Erforschung der PH-Domäne Leucin-reiche-Wiederholungs-Proteinphosphatase 2 (PHLPP2) haben anfängliche Studien von Gao et al. diese Phosphatase als Regulator des Protein-Kinase B (PKB, auch bekannt als Akt) Signalwegs identifiziert. Es wurde gezeigt, dass PHLPP2 eine Domänenstruktur aufweist, die eine Ras-assoziierte (RA) Domäne, eine Pleckstrin-Homologie (PH) Domäne, eine leucinreiche Wiederholungsdomäne (LRR), eine Protein-Phosphatase 2C (PP2C) Domäne und ein PDZ-Bindungsmotiv umfasst. Es wurde berichtet, dass sie spezifisch das regulatorische hydrophobe Motiv von Akt dephosphoryliert, was auf eine Rolle in den Zellüberlebens- und Wachstumspfaden hinweist. Jedoch legen umfassende Reinigungs- und biochemische Profilierungsstudien nahe, dass die beobachtete Phosphataseaktivität in früheren Studien von einer mitgereinigten Protein-Phosphatase 2A (PP2A), einer bekannten Akt-Phosphatase, und nicht von PHLPP2 selbst stammte. Der Einsatz von Okadasäure (OA), einem Inhibitor von PP2A, sowie die Entfernung von PP2A mit Phosphatase-Inhibitor-Perlen (PIBs), offenbarte keine intrinsische Phosphataseaktivität von PHLPP2 nach der Reinigung. Darüber hinaus haben wir experimentell bestimmt, dass das katalytische Domäne von PHLPP2 ein einzelnes Zn2+ Metallion verankert und wir präsentieren ein Modell für die Pseudo-Katalysestelle von PHLPP2 unter Verwendung von molekulardynamischen (MD) und quantenmechanischen (QM) Methoden. In Verbindung mit dem Fehlen von PHLPP2-Aktivität in Phosphatase-Assays stellen wir die Hypothese auf, dass PHLPP2 möglicherweise eine Pseudophosphatase ist, die unter den getesteten Bedingungen keine intrinsische enzymatische Aktivität aufweist. Diese Erkenntnis erfordert eine Neubewertung der Rolle von PHLPP2 und lädt zu rigoroseren experimentellen Ansätzen ein, um seine biologische Funktion zu klären.

In the exploration of PH domain Leucine-rich Repeat Protein Phosphatase 2 (PHLPP2), initial studies by Gao et al. identified this phosphatase as a regulator of Protein Kinase B (PKB, also known as Akt) signalling. PHLPP2 was shown to have a domain structure comprising a Ras-associated (RA) domain, pleckstrin homology (PH) domain, a leucine-rich repeat (LRR) domain, a Protein Phosphatase 2C (PP2C) domain, and a PDZ-binding motif. It was reported to specifically dephosphorylate the regulatory hydrophobic motif of Akt, suggesting a role in cell survival and growth pathways. However, comprehensive purification and biochemical profiling suggest that the observed phosphatase activity in earlier studies came from co-purified Protein Phosphatase 2A (PP2A), a known Akt phosphatase, rather than PHLPP2 itself. The use of okadaic acid (OA), an inhibitor of PP2A, as well as removal of PP2A with Phosphatase-Inhibitor Beads (PIBs), revealed no intrinsic PHLPP2 phosphatase activity post-purification. Furthermore, we experimentally determined that the catalytic domain of PHLPP2 anchors a single Zn2+ metal ion and we present a model for the pseudo-catalytic site of PHLPP2 using molecular dynamics (MD) and quantum mechanical (QM) methods. In conjunction with the lack of PHLPP2 activity in phosphatase assays, we raise the hypothesis that PHLPP2 may be a pseudophosphatase, lacking intrinsic enzymatic activity under the tested conditions. This insight necessitates a re-evaluation of PHLPP2's role, inviting more rigorous experimental designs to clarify its biological function.
