Laktose, eines der wesentlichsten Bestandteile der Molke, fällt bei der Herstellung von Milchprodukten als Abfallprodukt an. Vielversprechende Möglichkeiten zur Verwertung der Laktose bietet das Enzym PDH (Pyranose Dehydrogenase, EC 1.1.99.29). Ziel dieser Arbeit war die Charakterisierung rekombinanter AmPDH aus Aspergillus nidulans und aus Pichia pastoris, neuen Biokatalysatoren zur enzymatischen Herstellung von Laktulose und Laktobionsäure. Bei der Kultivierung der AmPDH aus Aspergillus nidulans konnten Ausbeuten von über 5 U/ml erreicht werden. Die Herstellung der rekombinanten AmPDH mit Pichia pastoris war besonders erfolgreich. Bei der Fermentation wurden bis zu 20 U/ml erzielt. Die anschließende Reinigung erfolgte in mehreren Schritten: Ammonsulfatfällung, Anionenaustauscherchromatographie, Hydrophobe Interaktionschromatographie und Gelfiltration. Die AmPDH aus Aspergillus nidulans zeigt ein Molekulargewicht von 67 kDa, die AmPDH aus Pichia pastoris ist mit 75 kDa etwas größer (ermittelt anhand der SDS- Page). Letztere weist auch einen höheren Zuckeranteil und deutlich mehr Isoformen auf. Beide rekombinanten PDHs zeichnen sich durch eine breite Substratspezifität aus. Bevorzugt werden Monosaccharide. Hinsichtlich der Gruppe der Elektronenakzeptoren hält sich die Auswahl bedingt. Es eignen sich Ferrocenium sowie einfache Chinone.

Lactose is among the most abundant carbohydrates. Its principal source is whey, a by-product in the dairy industry. New recycling processes and products based on lactose are under way to improve its total market. The enzyme PDH (pyranose dehydrogenase, EC 1.1.99.29) shows a great promise for carbohydrate conversions. The principle aim of this work was the characterization of two recombinant enzymes from two different sources, once from Aspergillus nidulans and once from Pichia pastoris for the enzymatic synthesis of lactulose and lactobionic acid. The cultivation of Aspergillus nidulans reached an enzyme- yield of about 5 U/ml, the fermentation of Pichia pastoris achieved 20 U/ml. Purification to a high homogeneity was obtained by ammonium sulphate precipitation, ion exchange chromatography, hydrophobic interaction chromatography and gelfiltration. The PDH was determind to be a glycosylated monomer. The Aspergillus nidulans AmPDH reveals a molecular weight of 67 kDa. The Pichia Pastoris AmPDH shows 75 kDa (reviewed by SDS- Page). The sugar content of the AmPDH from Pichia pastoris is higher than the sugar content for the Aspergillus nidulans AmPDH. Both enzymes show a broad sugar specificity. Monosaccharides are preferred substrates. Regarding the electron acceptors they are restricted. Ferrocenium and simple quinones are compatible.
