Krebs stellt eine aktuelle Bedrohung der öffentlichen Gesundheit dar, wobei die bestehenden Therapiemöglichkeiten noch Platz für Verbesserungen lassen. Der Grund dafür ist die Komplexität und Heterogenität dieser Krankheitsgruppe. Krebsentstehung und Wachstum ist nämlich tief verwurzelt in vielen biologischen Prozessen, welche aber nicht immer von den derzeitigen Behandlungen addressiert werden. Ein Faktor der zum Krankheitsverlauf beiträgt, ist die Tumormikroumgebung, in der verschiedene Zellarten Krebs auf unterschiedliche Art und Weise unterstützen können. Eine davon sind zum Beispiel die Tumor-assoziierten Makrophagen (TAM), die durch Funktionen des M2 Polarisierungszustands zur Bösartigkeit von Tumoren beitragen. Makrophagen können eine Vielzahl an biologischen Funktionen erf¨ullen durch Polarisierung zu verschiedenen Zuständen, beispielsweise die pro-entzündlichen M1 Makrophagen, die den anti-entzündlichen M2 Makrophagen gegenüberstehen. Es ist von Bedeutung die Mechanismen, die hinter der Makrophagenpolarisierung stecken, zu verstehen, um neue Krebstherapien zu entwickeln. Diese könnten nämlich die unterstützende Rolle der TAM addressieren, oder die antitumorale Eigenschaften des M1 Phänotyps in Immuntherapien anwenden. In dieser Arbeit wurden die Unterschiede zwischen den verschiedenen Polarisierungszuständen auf metabolischer und proteomischer Ebene untersucht, indem U937 Zellen zu M0-, M1- und M2 Makrophagen differenziert wurden mit anschließender LC-MS Analyse des Metaboloms und Proteoms. Zusätzlich wurde der Effekt von oxidativem Stress auf das Metabolom der unterschiedlich polarisierten Makrophagen untersucht, indem die Zellen mit tert-Butylhydroperoxid (TBHP) behandelt wurden. Reaktive Sauerstoffspezies spielen nämlich in den natürlichen Zellfunktionen von Makrophagen und in entzündetem Gewebe eine Rolle. Die LC-MS Analyse zeigte klare metabolische Unterschiede zwischen den verschiedenen Makrophagen-Typen, die mit der Expression von bestimmten Proteinen verifiziert wurden. Die Plastizität der Makrophagen wurde dadurch veranschaulicht, dass eine Mischung aus pro- und antientzündlichen Stoffwechselwegen in der gleichen Zelle gefunden wurden. Die TBHP Behandlung zeigte Auswirkung auf bestimmte Metabolite, nämlich Dihydroxyacetonphosphat, Fructose-1,6-bisphosphat, 3-Hydroxyanthranilsäure und Orotsäure.

Cancer is an ongoing detriment to public health with available treatment options still leaving much to be desired. The reason for this, is the complexity and heterogeneity of this disease group, with hallmarks spanning across multiple biological functions, which aren’t all addressed with current therapies. One of the factors playing into cancer progression is the tumor microenvironment (TME). In the TME there are multiple cell types that assist tumors in their growth, such as macrophages, called tumor associated macrophages (TAMs). These support cancer malignancy through pathways associated with the M2 macrophage polarization state. Macrophages can achieve a plethora of biological functions through polarization into different states, for instance the pro-inflammatory M1-like phenotype opposing the anti-inflammatory M2-like phenotype. Understanding the cellular polarization mechanisms is of importance to discover novel cancer treatments. These may address the supporting role of the TAMs in the TME or use the anti- cancer function of M1 macrophages as immunotherapy. In this work, the metabolic and proteomic signatures of macrophage polarization were studied by differentiating U937 cells into M0-, M1-, and M2 macrophages then subsequently analyzing the metabolome and proteome using LC-MS techniques. Additionally, the effect of an oxidative environment on the metabolome of the macrophages was investigated by treating the different cell types with tert-butyl hydroperoxide (TBHP), since reactive oxygen species (ROS) play a role in the cellular functions of macrophages as well as in inflamed tissue. The LC-MS analysis showed clear metabolic differences between the differently polarized macrophages that were confirmed with the expression profiles of respective proteins. The plasticity of macrophages was demonstrated by the fact that a mix of pro- and anti-inflammatory pathways were found in the same cell. Additionally, TBHP treatment had an impact on certain metabolites, namely dihydroxyacetone phosphate, fructose-1,6-biphosphate, 3-hydroxyanthranilic acid and orotic acid.
