Die Erforschung von Phosphonat (Pn) abbauenden Enzymen ist nach wie vor eine wissenschaftliche Niche, welche jedoch in den letzten Jahrzehnten vermehrt für Aufsehen gesorgt hat. Grund hierfür ist die Erkenntnis, dass es einen direkten Zusammenhang mit den biogeochemischen Zyklen der Natur (P, C und N-Zyklus) gibt. Da ein tieferes Verständnis des Pn-Abbaus zudem höchstwahrscheinlich zu medizinischen Innovationen führen wird, ist es notwendig Verbindungen herzustellen, welche als Grundlage für mechanistische Untersuchungen des Pn Metabolismus benötigt werden. Die gegenständliche Masterarbeit beschäftigt sich daher mit der Synthese diverse Substanzen für die Studien des Enzym Komplexes C−P Lyase sowie der Hilfsenzyme des hydrolytischen Pn-Abbauweges. Die Arbeit diskutiert die erfolgreiche Synthese von (2-(4-Nitrophenoxy)ethyl)phosphonsäure (NPEP), einen Substrat Analogon der C−P Lyase, welches als Indikator für C−P Lyase-aktivität in Umweltproben dienen soll. Weiters werden verschiedene Syntheseroute für (N-Dansyl-3-aminopropyl)2-tryptamine-ethylphosphinat (DTAP), welches in gleicher Art wie das zuvor erwähnte NPEP als C−P Lyase Indikator dienen soll, diskutiert. Zuletzt beschäftigt sich die Arbeit mit mehreren Versuchen beide Enantiomere von 2-Amino-1-fluoroethylphosphonsäure (FAEP) zu synthetisieren. FAEP ist ein potenzieller Inhibitor des Enzyms Phosphonate breakdown factor A (PbfA) welches jedoch über einen bislang unbekannten Mechanismus in Anwesenheit von Phosphonate breakdown factor F (PbfF) hydrolytisch gespalten werden kann.

Research on phosphonate (Pn) degrading enzymes is today still a scientific niche but received considerable attention within the past decades as its importance and relations to the major biogeochemical cycles (P, C, and N cycle) came to light. Studying the underlying mechanism of Pn breakdown furthermore has the potential to lead to significant medical innovations. Thus, synthetic efforts are needed to facilitate enzyme mechanistic studies on Pn degrading enzymes. The current thesis aimed at the synthesis of varied compounds needed to study open questions regarding the radical C−P lyase enzyme complex, as well as on auxiliary enzymes needed for hydrolytic Pn degradation. It presents the successful synthesis of (2-(4-nitrophenoxy)ethyl)phosphonic acid (NPEP), a C−P lyase substrate analogue which is planned to be used as indicator of C−P lyase activity in complex environmental samples. The publication further discusses varied synthetic approaches towards (N-dansy-3-aminopropyl)2-tryptamine ethyl phosphin (DTAP), which aims to detect C−P lyase activity in a similar way. Both compounds will facilitate the discovery of unstudied C−P lyase homologs and therefore help in investigating this breakdown mechanism. Additionally, attempts to access both enantiomers of 2-amino-1-fluoroethylphosphonate (FAEP) are described. This compound is a potential inhibitor of phosphonate breakdown factor A (PbfA), which can be degraded hydrolytically by an unknown mechanism in the presence of phosphonate breakdown factor F (PbfF). The latter is hypothesized to possess additional pharmaceutical value as a drug against phosphonate producing organisms.
