φCh1 ist ein temperenter Kopf-Schwanz-Virus, welcher N. magadii als einzigen bekannten Wirt verwendet. Der Wirt N. magadii, der im Sodasee Magadi beheimatet ist, lebt dort unter extremen Bedingungen, mit hohem pH-Wert und Salzgehalt und hat sich an diese Umgebung angepasst, was dieses Virus-Wirtspaar einzigartig und daher zu einem interessanten Studienobjekt macht. Viele Aspekte der Wechselwirkungen von φCh1 und N. magadii sind derzeit unbekannt oder nicht vollständig geklärt, so auch die Funktion der Open Reading Frames 80 und 82. Frühere Studien, die eine Sequenzanalyse und Vergleich durchführten, deuten darauf hin, dass es sich bei beiden ORFs wahrscheinlich um DNA-Methyltransferasen handelt, genauer bei ORF80 um eine m5C-mtase und bei ORF82 um eine m4C-mtase. Bei einem weiteren ORF, ORF94, wurde bereits bestätigt, dass es sich um eine Methyltransferase handelt, die eine wichtige Rolle bei der Regulation des viralen Lebenszyklus spielt. So wie ORF94 untersucht wurde, wurden nun auch in dieser Arbeit die Funktionen von ORF80 und ORF82 in φCh1 analysiert. Um dies zu erreichen, wurden erst Deletionsmutanten des lysogen infizierten Stammes von N. magadii, L11, erzeugt, indem jeweils einer oder beide ORFs durch eine Novobiocin-Resistenzkassette ersetzt wurden. Diese Deletionsmutanten wurden dann komplementiert, indem sie mit einem Plasmid transformiert wurden, welches die deletierten ORFs trug. Zusätzlich wurden Überexpressionsvarianten von L11 durch Transformation von regulärem L11 mit den Komplementationsplasmiden. erzeugt. Alle diese Konstrukte wurden dann hinsichtlich ihres Wachstumsverhaltens, ihres Virustiters und ihrer Produktionsmuster bestimmter Proteine von φCh1 verglichen.

φCh1 is a temperate head-tail virus, using N. magadii as it’s only known host. The host N. magadii, native to the soda lake Magadi, lives under extreme conditions of high pH and salinity and adapted to this environment, making this virus host pair somewhat unique and therefore an interesting subject of study. Many aspects of the interactions of φCh1 and N. magadii are currently unknown or not completely elucidated, which is also true for the function of the open reading frames 80 and 82. Previous studies, performing sequence analysis and comparison, indicate that both ORFs are likely to be DNA-methyl-transferases, particularly ORF80 seems to be a m5C-mtase and ORF82 a m4C-mtase. Another ORF, ORF94 was confirmed to be a methyltransferase, which plays a major role in the regulation of the viral lifecycle. Like ORF94 was studied before, so the functions of ORF80 and ORF82 in φCh1 were analysed in this thesis. To achieve this, first, deletion mutants of lysogenicly infected strains of N. magadii, L11, were created by replacing one or both ORFs with a novobiocin resistance cassette. These deletion mutants were then complemented by transforming them with a plasmid, carrying the deleted ORFs. Additionally, overexpression variants of L11 were created by transforming regular L11 with the complementation plasmids. All of these constructs were then compared in regard to their growth behaviour, viral-titers, and production patterns of certain proteins of φCh1.
