Die quantitative elementare Bildgebung biologischer Proben mittels Laserablation gekoppelt an Massenspektrometrie mit induktiv gekoppeltem Plasma (LA-ICP-MS) erfordert matrixangepasste Kalibrierstandards, die reproduzierbar, multielementfähig und mit Einzelzell-Hochdurchsatzanwendungen kompatibel sind. Gelatinebasierte Mikrotropfen-Standards wurden zuvor als matrixangepasste Referenzen für weiche biologische Gewebe validiert; ihre reproduzierbare Herstellung stellte jedoch einen praktischen Engpass für die routinemäßige Anwendung dar. Die vorliegende Arbeit adressiert diesen Engpass durch eine systematische Optimierung des gesamten Herstellungsprozesses – von der Dosier-hardware bis zur zerstörungsfreien Qualitätskontrolle. Zwei automatisierte Dosierplattformen wurden verglichen. Das pneumatische Certus-Flex-System wurde aufgrund säureinduzierter Metallkontamination an den Edelstahl-Mikroventilen sowie unkontrollierter Satellitentropfenbildung ausgeschlossen. Das piezoelektrische CellenONE-Mikrospottersystem wurde wegen seiner chemisch inerten Borosilikatglas-Kapillare und der reproduzierbaren Pikoliter-Dosierung weiterverfolgt. Ein Screening von sechs Biopolymermatrices über vier saure Lösungsmittelzusammensetzungen identifizierte 1 % (m/m) Kaltwasserfischgelatine (CWFSG) in 5 % (V/V) HNO3 als optimale Formulierung mit einer Tropfengenerierungsrate von 99.6 % bei minimaler feldinterner Streuung (RSDField = 5.7 %). Trocknungsinduzierte Inhomogenitäten infolge von Marangoni-Konvektion und Coffee-Ring-Effekt wurden durch Zusatz von 0.03 M Diglykolsäure (DGA) gemindert, die die zentrale Analytanreicherung unterdrückt, ohne Dosierzuverlässigkeit oder Tropfenmorphologie zu beeinträchtigen. Auf Basis eines Fluorescein-Tracers (25 μg mL−1) wurde eine fluoreszenzbasierte Vorabkontrolle entwickelt und gegen orthogonale ICP-MS-Messgrößen validiert: Die optische Tropfenfläche bildet die volumetrische Dosiervarianz präzise ab (RSD = 4.3 % to 4.4 %) und ermöglicht eine zerstörungsfreie Qualitätsbewertung jedes Tropfens vor der Laserablation. Die analytische Leistungsfähigkeit der finalen Formulierung wurde an einem Panel aus 48 Isotopen gegen eine Referenzcharge aus 2024 charakterisiert. Eine lineare Kalibrierung (R2 > 0,99) wurde für 43 der 48 Isotope erreicht; die methodischen Nachweisgrenzen erstrecken sich über sechs Größenordnungen – von sub-femtogramm-Werten für die schweren Lanthanoide (175Lu bei 0.026 fg pro abgetragener Region of Interest) bis zu zehntausenden Femtogramm für endogene Elemente mit hohem Untergrundsignal wie 23Na. Das resultierende Protokoll bildet eine reproduzierbare und übertragbare Grundlage für die routinemäßige quantitative LA-ICP-MS-Bildgebung sowie für künftige Ringversuchsstudien

Quantitative elemental bioimaging by laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) requires matrix-matched calibration standards that are simultaneously reproducible, multi-element capable, and compatible with high-throughput single-cell workflows. Gelatin-based microdroplet standards have previously been validated as matrix-matched references for soft biological tissue, but their reliable and reproducible production has remained a practical bottleneck that has limited their routine deployment. This thesis addresses that bottleneck through a systematic optimisation of the entire production workflow, from dispensing hardware to non-destructive quality control. Two automated dispensing platforms were compared. The pneumatically driven Certus Flex was disqualified on independent grounds of acid-induced metal leaching from its stainless-steel microvalves and uncontrolled satellite formation at its minimum nanoliter dispensing volume. The piezoelectric CellenONE micro-spotter was retained for its chemically inert borosilicate-glass capillary and its reproducible picoliter dispensing. A screen of six biopolymer matrices across four acidic solvent compositions identified 1 % (w/w) cold water fish skin gelatin (CWFSG) in 5 % (v/v) HNO3 as the formulation combining a droplet generation success rate of 99.6 % with the lowest intra-field variance (RSDField = 5.7 %). Drying-induced elemental inhomogeneities arising from Marangoni convection and the coffee-ring effect were mitigated by the addition of 0.03 M diglycolic acid (DGA), which suppresses central analyte accumulation without compromising spotting reliability or the macroscopic droplet morphology. A fluorescence-based pre-screening workflow employing a 25 μg mL−1 fluorescein tracer was developed and cross-validated against orthogonal ICP-MS readouts: the optical droplet area faithfully captured volumetric dispensing variance (RSD = 4.3 % to 4.4 %, in close agreement with the stabilised ICP-MS Signal Score), enabling per-droplet quality assessment of every printed array prior to laser ablation. The analytical performance of the finalised formulation was characterised across a 48-isotope element panel against a 2024 legacy reference. Linear calibration (R2 > 0.99) was achieved for 43 of the 48 evaluated isotopes, and methodological limits of detection span six orders of magnitude — from sub-femtogram values for the heavy lanthanides (175Lu at 0.026 fg per ablated region of interest) to tens of thousands of femtograms for high-background endogenous elements such as 23Na. The resulting protocol provides a reproducible and transferable basis for routine quantitative LA-ICP-MS bioimaging and for prospective interlaboratory comparison studies.
