Die meisten Antibiotika sind Naturstoffe, produziert von Mikroorganismen, die Schlüsselenzyme in fremden Organismen angreifen. Der übermäßige Einsatz von Antibiotika hat die Entwicklung von Resistenzmechanismen vorangetrieben und die Wirksamkeit vieler Antibiotika eingeschränkt. Daher müssen alternative Strategien zur Antibiotikaentwicklung erforscht und unser Verständnis von Resistenzmechanismen vertieft werden. Das erste Projekt untersuchte das Antibiotikaresistenzprotein CszB, welches Teil des Biosynthese-Genclusters in Xenorhabdus szentirmaii ist, das den DNA-Gyrase-Inhibitor Chloroszentirazine (Csz) herstellt. Da CszB ein Pentapeptide-Repeat-Protein (PRP) ist, wurde vermutet, dass CszB an die Gyrase von X. szentirmaii bindet, um diese zu schützen. Es wurde versucht, CszB im Komplex mit einem trunkierten Fusionskonstrukt der DNA-Bindungs-/Spaltungsdomäne der Gyrase von X. szentirmaii zu reinigen und zu kristallisieren. Da es keine Hinweise auf Komplexbildung gab, ist die Schlussfolgerung, dass CszB nicht an die DNA-Bindungs-/Spaltungsdomäne der Gyrase bindet. Das zweite Projekt erforschte, wie das Hybridantibiotikum Thiomarinol A die Isoleucyl-tRNA-Synthetase (IleRS) hemmt. Thiomarinol A vereint das IleRS-hemmende Antibiotikum Mupirocin mit einem Metall-chelatierenden Antibiotikum. Trotz der Ähnlichkeit zu Mupirocin bleibt Thiomarinol A wirksam gegen Mupirocin resistente Staphylococcus aureus Stämme. Durch biochemische Studien konnten wir eine starke Stabilisierung der Mupirocin empfindlichen IleRS1-Varianten durch Thiomarinol A demonstrieren, während die Mupirocin resistente IleRS2-Variante signifikant geringer stabilisiert wurde. Mit Cryo-Elektronenmikroskopie konnte eine vorläufige 3,5 Å auflösende Struktur von PmIleRS1 von Priestia megaterium gebunden an Thiomarinol A, erzielt werden. Diese Ergebnisse liefern die erste Struktur von IleRS gebunden an Thiomarinol A, und den ersten quantitativen Vergleich des Einflusses von Thiomarinol A auf IleRS1- und IleRS2-Varianten.

Most antibiotics are natural products evolved by microorganisms to target essential enzymes in other organisms. Extensive clinical use of antibiotics has driven resistance development, restricting many antibiotics’ effectiveness. Therefore, exploring alternative antibiotic development strategies and deepening our understanding of resistance mechanisms is crucial. The first topic of this thesis examines the putative self-resistance protein CszB, which is part of the biosynthetic gene cluster synthesizing the DNA gyrase inhibitor chloroszentirazine (Csz) in Xenorhabdus szentirmaii. As part of the pentapeptide repeat proteins (PRP), CszB is hypothesised to associate with X. szentirmaii’s gyrase, conferring self-resistance against Csz. Therefore, efforts were made to co-purify and co-crystallize CszB with a truncated fusion construct of the DNA-binding/cleavage domain of X. szentirmaii gyrase and investigate their interaction with biochemical methods. The results showed no evidence for complex formation, suggesting that CszB doesn’t associate with the DNA-binding/cleavage domain. The second project examines how the hybrid antibiotic thiomarinol A interacts with its target isoleucyl-tRNA synthetase (IleRS). Thiomarinol A combines an analogue of the IleRS-inhibiting antibiotic mupirocin with a metal-chelating antibiotic. Despite their similarities, thiomarinol A remains effective against high-level mupirocin-resistant Staphylococcus aureus. Through thermal shift assays, we demonstrated a strong stabilisation of mupirocin-susceptible IleRS1 variants, while the mupirocin-resistant IleRS2 variant was stabilised significantly less. Cryo-electron microscopy analysis produced a preliminary structure at 3.5 Å resolution of Priestia megaterium’s PmIleRS1, bound to thiomarinol A. These results provide the first thiomarinol A-bound IleRS structure and the first quantitative comparison of thiomarinol A’s interaction with IleRS1 and IleRS2 variants.
