In der Lebensmittelindustrie werden Milch und Sojagetränke aus Gründen der Produktsicherheit thermisch behandelt. Dieser Prozess kann bei verschiedenen Temperaturen durchgeführt werden. Dabei können Maillard-Reaktionsprodukte (MRPs) und Lipidoxidationsprodukte (LOPs) entstehen. Die Verdauung fördert deren weitere Bildung im Körper. Es ist jedoch unklar, wie sich die verschiedenen Erhitzungsmethoden für Milch und Sojagetränke auf diese Verbindungen während der Verdauung auswirken. Daher wurden in dieser Masterarbeit drei verschiedene wärmebehandelte Kuhmilchproben (LT-, HT- und UHT-Milch) sowie ein Sojagetränk ausgewählt und einer In-vitro-Verdauung unterzogen. Ziel war es, den Einfluss verschiedener thermischer Behandlungsmethoden auf die Bildung von MRPs und LOPs während der Verdauung zu bestimmen. Die mittels 1H-NMR nachgewiesene Lipidoxidation zeigte, dass unverdaute HT- und UHT-Milch (0,02 ± 0,01 mmol/mol Acylgruppen + Fettsäuren (AG+FA)) höhere n-Alkanal-Werte aufwiesen als LT-Milch. Die Verdauung förderte die Oxidation in allen Milchtypen (p < 0,05), verringerte jedoch die Unterschiede zwischen ihnen (0,14 ± 0,04 mmol/mol AG+FA). Generell wurde ein Anstieg der Lip-idoxidationsprodukte während der Verdauung beobachtet. Weitere mittels LC-QTOF bestimmte Lipidoxidationsprodukte, darunter oxidierte Triacylglycerine, Diacylglycerine, Monoacylglycerine und Fettsäuren, wurden in Kuhmilch, Sojamilch und deren Verdauungsprodukten identifiziert. Obwohl die Lipidoxidationsprodukte nach der Verdauung signifikant anstiegen, wurden für die meisten oxidierten Lipidarten keine signifikanten Unterschiede zwischen den verschiedenen verdauten Milchtypen festgestellt. Des Weiteren wurden neuartige oxidierte Lipide, insbesondere oxidierte DGs (z. B. DG 16:1_18:2;O₂ und DG 18:1_18:2;O) sowie ein oxidiertes MG (MG 18:3;O), identifiziert, was eine der wichtigsten Neuerungen dieser Arbeit darstellt. Im Hinblick auf die Maillard-Reaktion zeigte Furosin, ein frühes Maillard-Produkt, erhöhte Konzentrationen in unverdauter H-Milch. Nach der Verdauung wurde in allen Milchsorten und dem Sojagetränk ein signifikanter Anstieg der Gesamtkonzentration an AGEs beobachtet. Besonders hervorzuheben ist der Anstieg von MG-H1 (123367 ± 10112 µg/g Protein in HT-Milch und 134011 ± 5665 µg/g Protein in UHT-Milch) und G-H1 (82899 ± 2296 µg/g Protein in UHT-Milch und 80099 ± 3175 µg/g Protein in Sojadrink) nach der Verdauung. Da Vorstufen dieser MRPs über oxidative Wege gebildet werden können, deuten diese Ergebnisse darauf hin, dass eine gleichzeitige Lipidoxidation die Bildung von AGEs während der Verdauung fördern könnte. Im Einklang mit dem Ziel dieser Arbeit konnte eindeutig gezeigt werden, dass der Verdauungsprozess einen Anstieg von MRPs und LOPs verursacht. Insbesondere wurde der Gesamtanstieg freier MRPs sowie freier oxidierter Fettsäuren in den verdauten Proben zuverlässig charakterisiert und (semi-)quantifiziert. Diese Ergebnisse unterstreichen somit die Bedeutung der Untersuchung der Verdauungsdynamik von MRPs und LOPs nach Nahrungsaufnahme.

In the food industry, milk and soy beverages undergo thermal processing for product safety reasons. This process can be carried out at various temperatures. During this process, Maillard reaction products (MRPs) and lipid oxidation products (LOPs) can be formed. The digestive process pro-motes their further formation in the body. However, it is unclear how the different heating methods used for milk and soy beverages affect these compounds during digestion. Therefore, in this mas-ter’s thesis, three different heat-treated cow’s milk samples (LT, HT, and UHT milk) and one soy beverage were selected and subjected to in vitro digestion. Consequently, the objective was to de-termine the influence of different thermal processing methods on formation of MRPs and LOPs dur-ing digestion. Lipid oxidation, as detected by 1H-NMR, showed that undigested HT and UHT milk (0.02 ± 0.01 mmol/mol acyl groups + fatty acids (AG+FA)) exhibited higher n-alkanal levels compared to LT milk. Digestion promoted oxidation in all milk types (p < 0.05) but minimized the differences between them (0.14 ± 0.04 mmol/mol AG+FA). In general, an increase in lipid oxidation products during digestion was monitored. Other LOPs determined by LC-QTOF, including oxidized triacylglycerols, diacylglyc-erols, monoacylglycerols and fatty acids, were identified in cow milk, soy drink and their digestates. Although lipid oxidation products increased significantly after digestion, for most oxidized lipid spe-cies, no significant differences were observed among the various digested milk types. Furthermore, novel oxidized lipids, especially oxidized DGs (e.g. DG 16:1_18:2;O2 and DG 18:1_18:2;O) and one oxidized MG (MG 18:3;O), were identified, representing one of the main novelties of this thesis. Regarding the Maillard reaction, furosine, an early-stage Maillard product, showed elevated levels in non-digested UHT milk. A significant increase in the total AGE concentration was observed in all milk types and the soy beverage after digestion. Of particular note is the increase in MG-H1 (123367 ± 10112 µg/g protein in HT milk and 134011 ± 5665 µg/g protein in UHT milk) and G-H1 (82899 ± 2296 µg/g protein in UHT milk and 80099 ± 3175 µg/g protein in soy drink) after digestion. As pre-cursors for these MRPs can be generated via oxidative pathways, these findings indicate that simul-taneous lipid oxidation might promote the formation of AGEs during digestion. In line with the aim of this thesis, it was clearly demonstrated that the digestion process caused an increase in MRPs and LOPs. In particular, the summed increase in free MRPs as well as free oxi-dized fatty acids was reliably characterized and (semi)quantified in the digestate samples. Thus, these results highlight the importance of assessing digestive dynamics of MRPs and LOPs after food intake.
