(i) Erythropoiese benötig Signalübertragung über die Erythropoetin-Rezeptor (EpoR) Janus-Kinase 2 (Jak2) - Signal Transducer and Activator of Transcription 5 (Stat5) Achse, welche Proliferation, Differenzierung und Überleben von erythroiden Zellen steuert. Wir fanden heraus dass, Mäusen denen das Protein Stat5 fehlt, an mikrozytischer Anämie leiden aufgrund reduzierter Expression der anti-apoptotischen Proteine Bcl-xL und MCL-1, welches in erhöhter Apoptose erythroider Zellen resultiert. Zusätzlich sind die Zelloberflächenexpressionsmengen des Transferrinrezeptors-1 (TfR-1) auf erythroid Zellen von Stat5-/- Tieren, mehr als 2-fach reduziert. Dieser Rückgang wurde auf reduzierte Transkription der TfR-1 mRNA zusammen mit reduzierter Expression des Iron regulatory protein-2 (IRP-2) zurückgeführt. IRP2 ist das wichtigste Protein welches für die TfR-1 mRNA Stabilität in erythroid Zellen zuständig ist. Letztendlich zeigten wir, dass sowohl TfR-1 als auch IRP-2 direkte transkriptionelle Ziele von Stat5 sind. Damit wurde eine unerwartete mechanistische Verbindung zwischen dem EpoR/Jak/Stat Signalweg und Eisenstoffwechsel beschrieben, beides Prozesse die für Erythropoiese unerlässlich sind. (ii) Genetische Ablation des EpoR oder Jak2 führen zu embryonale Letalität infolge defekter Erythropoiese. Die unterschiedlichen Anteile EpoR/Jak2-induzierter Signalwege zur funktionellen Erythropoiese sind noch nicht völlig verstanden. Wir konnten zeigen, dass die Expression einer konstitutiv aktivierten Stat5a Mutante (cS5) ausreicht, um die Proliferationsdefekte von Jak2-/- und EpoR-/- Zellen in einer Epo-unabhängigen Art und Weise zu umgehen. Transplantation von Jak2-/- fötalen Leber Zellen, transduziert mit cS5, in bestrahlte Mäuse führte zu reifen erythroiden und myeloischen Zellen von Spenderherkunft bis zu 6 Monate nach der Transplantation. Zusammengefasst, konnten wir zeigen dass die Aktivierung von Stat5, ein entscheidender Effektorschritt unterhalb von Jak2 in Erythropoiese als auch in Myelopoiese darstellt. (iii & iv) Es wurde impliziert, dass Stat5 eine wichtige Rolle in der lymphoiden Entwicklung als auch leukämischen Transformation spielt. Die meisten Studien verwendeten jeodoch „Stat5-defiziente“ Mäuse, welche immer noch eine N-termiale trunkierte Version jenes Proteins exprämieren (Stat5deltaN/deltaN Mäuse). Wir analysierten die lymphoide Entwicklung von Stat5-/- Mäuse in denen der gesamte Stat5a/b Gen-Locus ablatiert wurde. CD8+-T-Lymphozyten, γδ T-Zellrezeptor-positive T-Zellen sowie CD4+ CD25+ FoxP3+ regulatorische T-Zellen waren völlig abwesend in Stat5-/- Tieren. Außerdem erwies sich FoxP3, ein Transkriptionsfaktor der unbedingt erforderlich für regulatorische T-Zell-Polarisation ist, als unmittelbares Ziel-Gen von Stat5. Des Weiteren, zeigten wir das B-Zell-Reifung in Stat5-/- Mäusen nicht über das Pre-Pro-B-Zell-Stadium möglich war. Am spannendsten jedoch war die Entdeckung dass, Stat5-/- Zellen gegenüber invitro-Transformation als auch in vivo Leukämieentwicklung, beides induziert durch Abelson Onkogene (bcr/abl p185 & v-abl), resistent waren.

(i) Erythropoiesis strictly depends on signal transduction through the erythropoietin receptor (EpoR) – Janus kinase 2 (Jak2) – signal transducer and activator of transcription 5 (Stat5) axis, regulating proliferation, differentiation and survival. Using mice completely lacking Stat5 we demonstrated that these animals suffer from microcytic anemia due to reduced expression of the anti-apoptotic proteins Bcl-xL and Mcl-1 followed by enhanced apoptosis. Moreover, transferrin receptor-1 (TfR-1) cell surface levels were decreased >2-fold on erythroid cells of Stat5-/- animals. This reduction could be attributed to reduced transcription of TfR-1 mRNA and iron regulatory protein 2 (IRP-2), the major translational regulator of TfR-1 mRNA stability in erythroid cells. Both genes were demonstrated to be direct transcriptional targets of Stat5. This established an unexpected mechanistic link between EpoR/Jak/Stat signaling and iron metabolism, processes absolutely essential for erythropoiesis and life. (ii) Deletion of EpoR or Jak2 causes embryonic lethality as a result of defective erythropoiesis. The contribution of distinct EpoR/Jak2-induced signaling pathways to functional erythropoiesis is incompletely understood. We demonstrated that expression of a constitutively activated Stat5a mutant (cS5) was sufficient to relieve the proliferation defect of Jak2-/- and EpoR-/- cells in an Epo independent manner. Transplantation of Jak2-/- fetal liver cells transduced with cS5, into irradiated mice gave rise to mature erythroid and myeloid cells of donor origin up to 6 months after transplantation. In conclusion, we demonstrated that activated Stat5 is a critical downstream effector of Jak2 in erythropoiesis/myelopoiesis. (iii & iv) Stat5 has been implicated in lymphoid development and transformation. Most studies have employed Stat5-deficient mice where gene targeting only disrupted the first protein-coding exon, resulting in expression of N-terminally truncated forms of Stat5 (Stat5deltaN/deltaN mice). We reanalyzed lymphoid development in Stat5-/- mice having a complete deletion of the Stat5a/b gene locus. CD8+-T-lymphocytes, γδ T-cell receptor positive T-cells as well as CD4+ CD25+ FoxP3+ regulatory T-cells were completely absent in Stat5-/- animals. Furthermore FoxP3, the transcription factor strictly required for regulatory T-cell polarization was shown to be a direct target gene of Stat5. Additionally, B-cell maturation was abrogated at the pre–pro-B-cell stage in Stat5-/- mice. Most strikingly, however, Stat5-/- cells were resistant to in vitro transformation and in vivo leukemia development induced by Abelson oncogenes (bcr/abl p185 & v-abl).
